摘  要:目的:分析和表征圣愈汤干预骨髓抑制模型大鼠的入血成分,为其药效物质基础研究及质量控制提供依据。方法:利用大鼠体质量、血常规临床指标评价圣愈汤给药的有效性。以骨髓抑制SD大鼠为试验对象,灌胃给予圣愈汤,在显效状态下收集血浆样品,利用超高效液相色谱-四极杆-飞行时间串联质谱(UPLC-Q-TOF-MS)技术结合UNIFI系统分析表征吸收入血的原型成分及其代谢产物。结果:圣愈汤能够显著增加骨髓抑制大鼠的体质量、血红蛋白(HB)、红细胞计数(RBC)、血小板计数(PLT);在显效状态下,共表征出圣愈汤入血原型成分26个,代谢产物成分20个。主要血中移行成分包括梓醇、川芎内酯A、阿魏酸、芒柄花素、毛蕊异黄酮、人参皂苷Rb;、人参皂苷Rg;、人参皂苷Rg;、人参皂苷Rg;以及黄芪甲苷的代谢产物,具有对骨髓抑制的干预作用,为关键的活性成分。结论:研究结果初步阐释了圣愈汤干预骨髓抑制的药效物质基础,为圣愈汤药效物质研究和质量控制标准的提升提供科学依据。Objective:To analyze and characterize the plasma components of Shengyu Decoction(圣愈汤)in the intervention of bone marrow suppression in rats to provide a basis for its pharmacodynamic material basis research and quality control.Methods:The body weight and blood routine indexes of rats were measured to evaluate the effectiveness of Shengyu Decoction.SD rats with bone marrow suppression were administered with Shengyu Decoction by gavage.Plasma samples were collected under the effective state.Ultra high performance liquid chromatography quadrupole time of flight tandem mass spectrometry(UPLC-Q-TOF-MS)combined with UNIFI system was used to analyze and characterize the prototype components and metabolites absorbed into the blood.Results:Shengyu Decoction significantly increased the body weight,hemoglobin(HB),red blood cell count(RBC),and platelet count(PLT)of rats with bone marrow suppression.Twenty-six prototype components and 20 metabolites were characterized under the effective state.The main plasma components included catalpol,cnidilide,ferulic acid,formononetin,calycosin,ginsenoside Rb1,ginsenoside Rg1,ginsenoside Rg2,ginsenoside Rg3 and astragaloside IV metabolites,which showed intervention effect on bone marrow inhibition and were the key active components.Conclusion:The present study preliminarily explained the pharmacodynamic material basis of Shengyu Decoction in the intervention of bone marrow suppression and provided a scientific basis for the research of pharmacodynamic materials of Shengyu Decoction and the improvement of quality control standards.
