摘  要:目的:建立心肾阳虚型慢性心衰(CHF)大鼠模型,探讨符合中医证候理论的动物模型的建立标准。方法:动物分为3组,假手术对照组、模型对照组及参附汤3.38 g/kg组,造模组尾静脉注射阿霉素并切除甲状腺构建心肾阳虚心衰模型,对比心衰模型在血流动力学、病理及体征方面的变化探讨病证模型的特点,并通过参附汤验证模型。结果:与假手术对照组相比,模型对照组大鼠自然情况表现较差,体质量、体温显著降低,LVSP、+dp/dt_(max)、HR、LVEF显著降低(P<0.01),血清BNP含量显著升高(P<0.01),病理及透射电镜显示模型对照组大鼠心肌损伤明显;与模型对照组相比,参附汤3.38 g/kg组大鼠自然状态改善,体质量增加、体温明显升高(P<0.05),LVSP、+dp/dt_(max)、HR、LVEF显著升高(P<0.01),血清BNP含量显著降低(P<0.01),病理及透射电镜显示大鼠心肌损伤明显改善。结论:本研究建立的心肾阳虚型CHF大鼠模型,符合中医辨证及现代医学病证特点,为探索心肾阳虚病证结合的动物模型制作及评价提供了依据。Objective:To study the establishment of the rat model of chronic heart failure(CHF)with Xinshenyangxu(心肾阳虚)syndrome and to explore the criteria for the establishment of animal models in accordance with the syndrome theory in traditional Chinese medicine(TCM).Methods:Rats were divided into control group,model group,and Ginseng and Aconiti Praeparatae Decoctiongroup(3.38 g/kg).The CHF modelwith Xinshenyangxu syndrome was established by tail intravenous injection of adriamycin and removal of the thyroid gland.The characteristics of the CHF model were discussed in terms of hemodynamics,pathological sections,and signs,and the model was verified by Ginseng and Aconiti Praeparatae Decoction.Results:Compared with the control group,the model group performed poorly in natural conditions,and showed decreased body weight and body temperature,reduced left ventricular systolic pressure(LVSP),+dp/dt_(max),heart rate(HR),and left ventricular ejection fraction(LVEF)(P<0.01),and increased serum brain natriuretic peptide(BNP)level(P<0.01),with obvious myocardial damage by pathology and transmission electron microscopy.Compared with the model group,the Ginseng and Aconiti Praeparatae Decoction group(3.38 g/kg)demonstrated improved natural state,raised body weight and body temperature(P<0.05),increased LVSP,+dp/dt_(max),HR,and LVEF(P<0.01),and decreased serum BNP level(P<0.01),with alleviated myocardial injury by pathology and transmission electron microscopy.Conclusion:The CHF rat model with Xinshenyangxu syndrome was established by combining TCM syndrome differentiation and modern biological techniques,which provides a basis for the establishment and evaluation of animal models with Xinshenyangxu syndrome.
