墓头回水提液凉血止血作用及机制评价研究

摘　　要：目的:通过体内实验研究及网络药理学探究墓头回水提液对干酵母联合无水乙醇所致血热出血大鼠模型和正常小鼠凝血系统的作用,探究墓头回水提液的止血作用及其作用机制。方法:断尾法和毛细管法观察小鼠出血和凝血时间。自动凝血分析仪检测干酵母所致血热出血大鼠凝血四项指标;全自动血液细胞分析仪检测干酵母所致血热出血大鼠血常规指标;通过网络药理学筛选墓头回止血的活性成分、作用靶点及通路,并通过分子对接进行验证。结果:墓头回水提液5、10 g/kg组均能显著缩短小鼠出血时间(P<0.05或P<0.01),墓头回水提液5、10、20 g/kg组均能显著缩短小鼠凝血时间(P<0.01);与正常对照组比较,模型对照组大鼠的凝血酶原时间(PT)、凝血活酶时间(APTT)和血浆凝血酶时间(TT)均显著延长(P<0.01),纤维蛋白原(FIB)均显著增加(P<0.01),血小板计数(PLT)和血小板压积(PCT)显著减少(P<0.05或P<0.01),平均血小板体积(MPV)、血小板分布宽度(PDW)、血红蛋白量(HGB)、平均红细胞血红蛋白浓度(MCHC)、红细胞计数(RBC)、红细胞压积(HCT)均明显增加(P<0.05或P<0.01)。与模型对照组比较,墓头回水提液5、10 g/kg组均能减少血热出血大鼠TT、APTT、PT和FIB(P<0.05或P<0.01);除墓头回5 g/kg大鼠PDW有所减少但无统计学意义以外,各剂量墓头回水提液均能显著增加血热出血大鼠PLT和PCT(P<0.01),减少MPV和PDW(P<0.01);墓头回水提液10、20 g/kg组均能降低血热出血大鼠RBC、HCT、MCV、HGB(P<0.01),墓头回5 g/kg能显著降低RBC和HCT(P<0.01)。通过网络药理学筛选出邻苯二甲酸二异丁酯、槲皮素、邻苯二甲酸二丁酯、顺式十八烯-9-酸、反亚油酸甲酯、β-古芸烯和α-古芸烯7个墓头回止血活性成分,STAT3、MMP9、MPO、CCR5和JAK25个止血核心靶点,涉及神经活性配体-受体相互作用信号通道、钙信号通路、花生四烯酸代谢信号通道、趋化因子信�Objective:To examine the effect of aqueous extract of Mutouhui(墓头回)on coagulation systems in rats with Xuerechuxue(血热出血)syndrome induced by dry yeast and absolute ethanol and normal mice by in vivo experiment and network pharmacology and thereby investigate its hemostatic effect and the mechanism.Methods:With the tail-breaking method and capillary tube method,the bleeding time and clotting time of mice was determined.Four coagulation indicators of the rats with Xuerechuxue(血热出血)syndrome were detected by an automatic coagulation analyzer,and the routine blood indexes by an automatic hematology analyzer.The hemostatic components of Mutouhui(墓头回),and related targets and pathways were screened by network pharmacology and validated by molecular docking.Results:The bleeding time of mice was significantly shortened in the 5 g/kg and 10 g/kg Mutouhui(墓头回)groups(P<0.05 or P<0.01),and the clotting time was significantly reduced in 5,10,and 20 g/kg Mutouhui(墓头回)groups(P<0.01).Compared with normal control rats,the model control group showed longer prothrombin time(PT),partial thromboplastin time(APTT),and thrombin time(TT)(P<0.01),increase of fibrinogen(FIB)(P<0.01),decrease of platelet count(PLT)and plateletcrit(PCT)(P<0.05 or P<0.01),and rise of mean platelet volume(MPV),platelet distribution width(PDW),hemoglobin(HGB),mean corpuscular hemoglobin concentration(MCHC),red blood cell count(RBC),and hematocrit(HCT)(P<0.05 or P<0.01).Compared with model control rats,5 and 10 g/kg Mutouhui(墓头回)reduced TT,APTT,PT,and FIB in rats with Xuerechuxue(血热出血)syndrome(P<0.05 or P<0.01).Except that the reduction of PDW in the 5 g/kg Mutouhui(墓头回)group was insignificant,the aqueous extract of Mutouhui(墓头回)at each dose increased PLT and PCT(P<0.01),and decreased MPV and PDW(P<0.01)in model rats.The RBC,HCT,MCV,and HGB were significantly decreased in 10 g/kg and 20 g/kg Mutouhui(墓头回)groups(P<0.01),while RBC and HCT were significantly reduced in the 5 g/kg Mutouhui(墓头�
