摘 要:目的探究黄芩片通过调控NLRP3/Caspase-1通路影响大鼠肺炎呼吸衰竭的分子机制。方法购买SD健康雄性大鼠,尾静脉注射脂多糖(0.2 mg/100 g)构建呼吸衰竭模型,分为Normal组、Model组和黄芩片组,各6只。检测各组大鼠呼吸频率、氢离子浓度(pH)、二氧化碳分压(PaCO_(2))、氧分压(PO_(2))、50%用力呼气流量(FEF50)和呼气峰流速值(PEF)水平,HE染色观察肺组织病理形态学,ELISA检测炎症因子IL-1β、IL-8、IL-10和IL-17表达水平,qRT-PCR和Western Blot检测NLRP3和Caspase-1的表达水平。结果Normal组肺泡结构完整,大小均匀,Model组肺泡间隔增加,发现炎性细胞浸润,而黄芩片组炎性细胞浸润程度下降;与Model组相比,黄芩片组的呼吸频率、pH、FEF50、PEF、PaCO_(2)和PaO_(2)显著降低(P<0.05),炎症因子IL-1β、IL-8、IL-10和IL-17表达水平显著降低(P<0.05),和Model相比,黄芩片组的NLRP3和Caspase-1的mRNA和蛋白表达水平均有所降低(P<0.05)。结论黄芩片通过可能通过调控NLRP3/Caspase-1通路影响肺炎呼吸衰竭大鼠的炎症反应和疾病进程。Objective To explore the molecular mechanism of Huangqin Tablet(黄芩片)affecting pneumonia respiratory failure rats by regulating the NLRP3/Caspase-1 pathway.Methods SD healthy male rats were purchased and the respiratory failure model was established by tail vein injection of lipopolysaccharide(0.2 mg/100 g).They were divided into the control group,the model group and the Huangqin Tablet group,with 6 rats in each group.The respiratory rate,the hydrogen ion concentration(pH),the levels of partial pressure of carbon dioxide(PaCO_(2)),partial pressure of oxygen(PO_(2)),50%forced expiratory flow(FEF50)and peak expiratory flow rate(PEF)were measured.The pathomorphology of lung tissue was observed by HE staining,and the inflammatory factor IL-1βwas detected by ELISA.The expression levels of IL-8,IL-10 and IL-17 were detected by QRT PCR and Western Blot.Results In the normal group,the alveolar structure was complete and the size was uniform.In the model group,the alveolar septum was increased and the inflammatory cell infiltration was found.While in the Huangqin Tablet group,the degree of inflammatory cell infiltration was decreased.Compared to those of the model group,the respiratory rate,the values of pH,FEF50,PEF,PaCO_(2) and PaO_(2) in the Huangqin Tablet group were significantly lower(P<0.05),and the expression levels of inflammatory factors IL-1β,IL-8,IL-10 and IL-17 were decreased significantly(P<0.05).Compared with those of the model group,the mRNA and protein expression levels of NLRP3 and Caspase-1 in the Huangqin Tablet group were decreased(P<0.05).Conclusion Huangqin Tablet may affect the inflammatory response and the disease process of rats with pneumonia respiratory failure through regulating the NLRP3/Caspase-1 pathway.
