摘  要:为探讨硒代半胱氨酸(Sec)在镉(Cd)致成骨细胞凋亡中的保护作用,用20μg/m L Se-(甲基)硒代半胱氨酸盐酸盐预处理ROS1728细胞24 h后,添加20μg/m L氯化镉处理6 h。利用流式细胞术检测细胞凋亡率、线粒体膜电位(MMP)、活性氧簇(ROS);高内涵细胞成像系统分析细胞线粒体纹理指数;Western-blot法检测Bax和Bcl-2蛋白表达。结果显示,Sec能极显著抑制Cd引起的ROS1728细胞凋亡率的上升,极显著抑制Bax/Bcl-2比值的增高、MMP的下降、线粒体纹理结构指数的下降及ROS含量的升高(P<0.01)。结果表明,Sec通过降低ROS1728细胞内过氧化物含量,维持线粒体功能和结构完整,抑制Cd诱导的ROS1728细胞凋亡,为临床应用Sec防治Cd暴露引起的氧化应激反应导致的骨骼疾病提供理论指导。To investigate the protective effect of selenocysetine(Sec)on cadmium(Cd)-induced apoptosis of ROS1728 cells,ROS1728 cells were pretreated with 20μg/m L Se-(Methyl)selenocysteine hydrochloride for 24 h,and then exposed to 20μg/m L Cd Cl2for 6 h.Flow cytometry was applied to detect cell apoptotic rate,mitochondrial membrane potential(MMP),and reactive oxygen species(ROS).The expression of Bax and Bcl-2 was measured by Western-blot.High Content Analysis System(HCAS)was used to observe mitochondrial structure and analyze the index of mitochondrial structure.The results of this study showed that Sec significantly alleviated Cd-induced the increase of apoptotic rate(P<0.01),rise of Bax/Bcl-2 rate,decrease of MMP,reduction of mitochondrial structure index,and the accumulation of ROS content in ROS1728 cells.Our results showed that Sec inhibits Cd-induced apoptosis of ROS1728 cells by maintaining the function and intact structure of mitochondria via reducing ROS content in ROS1728cells.Our results would provide theoretical guidance for clinical application of Sec to pretreat Cdinduced bone metabolic diseases.
