摘 要:为筛选狮头鹅17β-羟基类固醇脱氢酶Ⅱ基因(17β-HSD2)的SNPs位点并进行生物信息学分析,采用直接测序法检测17β-HSD2基因外显子5区域的SNPs位点,并利用生物信息学软件分析突变前后mRNA二级结构的变化。结果表明,在狮头鹅群体中共发现6个SNPs,仅Exon5-351 G>A为错义突变。17β-HSD2基因突变前后mRNA最小自由能(MFE)、质心二级、含有假结的mRNA二级与折叠结构,均发生变化,且MFE自由能上升,结构稳定性变弱。结果可为后续探究17β-HSD2基因对狮头鹅繁殖性状的影响机制提供基础数据。In order to analyze the SNPs sites of 17β-Hydroxysteroid dehydrogenaseⅡ(17β-HSD2 gene)in Shitou goose,bioinformatics,SNPs in exon5 of 17β-HSD2 gene were detected by direct sequencing,and the changes of mRNA secondary structure before and after mutation were analyzed by bioinformatics software.The results showed that six SNPs were found in the Shitou goose population,and only exon5-351 G>A was a missense mutation.Before and after 17β-HSD2 gene mutation,the mRNA minimum free energy(MFE),centroid secondary,mRNA secondary and folding structure containing pseudoknots changed,and the MFE free energy increased and the structural stability weakened.The results could provide basic data for the follow-up study on the influence mechanism of 17β-HSD2 gene on reproductive traits of Shitou geese.
