摘  要:目的 探究柠檬醛(citral)抗限制性马拉色菌活性及其在豚鼠皮肤感染模型中的止痒作用。方法 采取微量倍比稀释法测定柠檬醛对限制性马拉色菌的最低抑菌浓度(minimum inhibitory concentration, MIC);建立限制性马拉色菌致豚鼠皮肤感染模型,分别从皮损评分、真菌转阴率、组织病理学、免疫组织化学Ki67四方面综合评价柠檬醛对限制性马拉色菌所致豚鼠皮肤感染的作用;建立组胺致豚鼠皮肤瘙痒模型,探讨柠檬醛的止痒作用。结果 柠檬醛对限制性马拉色菌的MIC值为(0.14±0.076)μg/mL。与正常组比较,模型组和空白基质组豚鼠皮肤均可见角质层破损严重、表皮角质化过度、颗粒层和棘层增厚等症状。给药7 d后,与模型组和空白基质组比较,各给药组豚鼠上述症状均有不同程度的减轻,其中柠檬醛中、高剂量组可显著降低豚鼠感染模型皮损评分(P<0.05);柠檬醛低、中、高剂量组的真菌转阴率分别为37.50%、50.00%、62.50%,免疫组织化学染色结果表明柠檬醛中、高剂量组可抑制表皮增殖因子Ki67的阳性表达;同时柠檬醛中、高剂量组可提高豚鼠的致痒阈值(P<0.05)。结论 柠檬醛具有较好的抗限制性马拉色菌活性,在马拉色菌所致豚鼠皮肤感染模型中有较好的止痒作用。Objective To investigate the antibacterial activity of citral on Malassezia restricta and its antipruritic effect in guinea pig skin infection model.Methods The minimum inhibitory concentration(MIC) of citral against Malassezia restricta was determined by microfold dilution method. A model of skin infection caused by Malassezia restricta was established, and the effects of citral on skin infection caused by Malassezia restricta were evaluated in terms of lesion score, fungal transfer rate, histopathology and immunohistochemistry Ki67. A model of histamine-induced pruritus in guinea pigs was set up to investigate the antipruritic effect of citral.Results The MIC value of citral against Malassezia restricta was(0.14±0.076)μg/mL. Compared with the normal group, severe cuticle breakage, hyperkeratinization of the epidermis, and thickening of the granular and spiny layers were seen in the skin of guinea pigs in both the model and blank substrate groups. After 7 days of administration, compared with the model group and the blank matrix group, the above symptoms were reduced to different degrees in each administration group, among which the citral middle and high dose groups could significantly reduce the skin lesion score of the guinea pig infection model(P<0.05);the fungal conversion rates of the citral low, middle and high dose groups were 37.50%, 50.00% and 62.50%, respectively, and immunohistochemical staining results showed that citral inhibited the positive expression of epidermal proliferation factor Ki67 in the middle and high dose groups;meanwhile, the middle dose and high dose of citral could improve the itching threshold of guinea pigs(P<0.05).Conclusion Citral has excellent antifungal activity against Malassezia restricta, and has a good antipruritic effect in guinea pig skin infection model caused by Malassezia restricta.
