摘 要:目的建立双波长HPLC法测定清喉咽颗粒中的黄芩苷、连翘苷含量。方法Agilent C_(18)色谱柱(4.6 mm×250 mm,5μm);流动相:乙腈-0.1%磷酸溶液(30∶70);检测波长:278 nm(黄芩苷),205 nm(连翘苷);柱温:30℃;流速:1 mL·min^(-1),进样量5μL。结果黄芩苷、连翘苷对照品溶液分别在0.06635~1.9906,0.006116~0.2447μg的质量范围内与其峰面积均呈良好的线性关系(r=1,0.9999);黄芩苷、连翘苷平均加样回收率分别为96.30%,97.03%(n=6)。结论所建立的测定方法操作简便、重复性好,可用于清喉咽颗粒中的黄芩苷、连翘苷的含量测定。OBJECTIVE To develop a dual wavelength HPLC method for simultaneously determination of baicalin and forsythin in Qinghouyan granules.METHODS Agilent C_(18)(4.6 mm×250 mm,5μm)column was used.The mobile phase was acetonitrile-0.1%phosphate(30∶70).The column temperature was 30℃.The detection wavelength was 278 nm(baicalin),205 nm(forsythin).RESULTS The linear ranges of baicalin and forsythin were between 0.06635-1.9906μg·mL^(-1)(r=1),0.006116-0.2447μg·mL^(-1)(r=0.9999).The average recoveries(n=6)were 96.30%,97.03%,respectively.CONCLUSION The established method is simple and reproducible.It can be used for the determination of baicalin and forsythin in Qinghouyan granules.
