摘 要:目的探讨白桦脂醇对慢性铝暴露诱导痴呆大鼠认知与情绪的干预作用,以及其对杏仁核细胞因子的影响机制。方法将无特定病原体级健康雌性SD大鼠随机分为对照组、模型组、干预对照组、低剂量组和高剂量组,每组10只大鼠;后4组大鼠均予剂量为100 mg/kg体质量的质量浓度为10.0 g/L氯化铝溶液灌胃,对照组大鼠予等体积蒸馏水灌胃,每天1次,连续60 d,建立慢性铝暴露致痴呆大鼠模型。干预对照组大鼠予质量浓度为0.5 g/L的盐酸多奈哌齐灌胃,低、高剂量组大鼠分别予质量浓度为20.0、60.0 g/L的白桦脂醇水溶液灌胃,对照组和模型组大鼠均予等体积蒸馏水灌胃,每天1次,连续4周。采用Y迷宫新异臂实验和高架十字迷宫实验评估大鼠焦虑样行为,采用强迫游泳实验评价大鼠抑郁样行为;采用免疫组化法检测大鼠杏仁核中磷酸化的Tau(p-Tau)蛋白和淀粉样前体蛋白(βAPP)相对表达水平;采用酶联免疫吸附测定法检测大鼠杏仁核中白细胞介素(IL)-1、IL-6和IL-8相对表达水平,采用蛋白质印迹法检测大鼠杏仁核中基质细胞衍生因子-1(SDF-1)、趋化因子受体(CXCR)3、CXCR4、干扰素诱导蛋白(IP)-10相对表达水平。结果行为学实验中,与对照组比较,模型组大鼠在新异臂中停留时间和开臂内停留时间百分比均减少(P值均<0.05),累计不动时间增加(P<0.05);与模型组比较,干预对照组、低剂量组和高剂量组大鼠在新异臂中停留时间和开臂内停留时间百分比均增加(P值均<0.05),累计不动时间均减少(P值均<0.05);与干预对照组比较,低剂量组和高剂量组大鼠在新异臂中停留时间和开臂内停留时间百分比均增加(P值均<0.05),累计不动时间均减少(P值均<0.05);与低剂量组比较,高剂量组大鼠在新异臂中停留时间和开臂内停留时间百分比均增加(P值均<0.05),累计不动时间减少(P<0.05)。与对照组比较,模型组大鼠杏仁核中p-TObjective To investigate the effect of betulin on the cognition and emotion of rats with chronic aluminum-induced dementia and its influence on the expression of inflammatory factors in amygdala.Methods The specific pathogen free female SD rats were randomly divided into control group,model group,intervention control group,low-dose group and high-dose group,with 10 rats in each group.The rats in the later four groups were given with 100 mg/kg body weight and 10.0 g/L aluminum chloride solution by gavage,while the rats in the control group were gavaged with the same volume of distilled water,once a day for 60 days.The rat model of dementia caused by chronic aluminum exposure was established.The rats in the intervention control group were given donepezil hydrochloride(0.5 g/L)by gavage,the rats in the low-dose and high-dose groups were given betulin(20.0 and 60.0 g/L)by gavage,and the rats in the control and model groups were given the same volume of distilled water by gavage,once a day for four weeks.Anxiety-like behavior was evaluated by Y maze new arm test and elevated cross maze test.The depression-like behavior was evaluated by forced swimming test.The amygdala of rats was isolated,and the expression of phosphorylated Tau(p-Tau)protein andβ-amyloid precursor protein(βAPP)was detected by immunohistochemistry.The expression of interleukin(IL)-1,IL-6 and IL-8 was detected by enzyme-linked immunosorbent assay.The expression of stromal cell-derived factor-1(SDF-1),C-X-C chemokine receptor(CXCR)3,CXCR4 and interferon-inducible protein-10(IP-10)was detected by Western blotting.Results In the behavioral experiments,the residence time in the new arm and the percentage of time in the open arm of the model group decreased(all P<0.05),while the cumulative immobility time increased(P<0.05)compared with the control group.The residence time in the new arm and the percentage of time in the open arm increased in the intervention control group,low-dose group and high-dose group(all P<0.05),while the cumulative immobility time
