摘 要:目的筛选制备硫酸黄连素多囊脂质体的稳定剂,制备得到稳定性好,大小均匀的硫酸黄连素多囊脂质体。方法采用二次复乳法制备硫酸黄连素多囊脂质体;以制得复乳是否凝集或出现凝集现象的时间长短、囊泡的稳定性为指标,考察不同稳定剂对多囊脂质体制备的影响。结果最适外水相稳定剂为海藻酸钠,采用优选处方制备硫酸黄连素多囊脂质体可以形成稳定复乳,并在去除有机溶剂前后都能在显微镜看到大小均匀的圆整有膜囊泡。结论以海藻酸钠为稳定剂,采用二次复乳法可以制得稳定、均匀的硫酸黄连素多囊脂质体。Objective To select the stabilizing agents to prepare stable berberine sulphate multivesicular liposomes with uniform sizes.Methods The berberine sulphate multivesicular liposomes were prepared via multiple emulsion method.The influence of different stabilizing agents on the preparation was studied through the observation of agglutination,the measurement of the agglutination time,and the determination of the vesicle stability.Results The suitable stabilizing agent in the outer solution was sodium alginate.With optimized formula,stable multiple emulsion of berberine sulphate multivesicular liposomes were successfully prepared.Micro vesicles with uniform size and intact membrane could be observed under microscope before and after the elimination of organic solvent.Conclusion Using sodium alginate as the stabilizing agent,stable berberine sulphate multivesicular liposomes with uniform sizes could be prepared via multiple emulsion method.
