摘 要:目的鉴定cisAB血型,了解其血清学及基因型特点,探讨cisAB血型患者输血策略。方法采用血型血清学方法检测5例标本的ABO血型;运用特异性序列引物聚合酶链反应技术进行ABO和cisAB血型基因分型;采用序列测定分型技术对3例ABO亚型样本进行1-7号外显子基因序列分析。结果样本1血清学分型为A_(2)Bx,基因分型为cisAB01/O02,样本2、样本3的血清学分型为A_(2)B,基因分型为cisAB01/B101,样本4血型为AB型,样本5血型为A型。1-3号样本的ABO血型基因分型结果与血清学结果不一致。结论cisAB01与不同ABO血型等位基因单倍型组合时,其血清学结果不尽相同,采用分子生物学检测技术对于辅助准确鉴定cisAB血型,选择合适的血液用于临床输注,保障输血安全具有重要意义。Objective To identify the cisAB blood group,to describe its serological and genotype characteristics,and to explore blood transfusion strategy for patients with cisAB blood group.Methods Blood group serological test was used to detect ABO blood group in 5 samples.Polymerase chain reaction with sequence specific primers(PCR-SSP)was employed to perform ABO and cisAB blood group genotyping.Exon 1-7 of the ABO genes in 3 samples were tested by sequencing based typing(SBT).Results The phenotype of sample 1 was A_(2)Bx and the genotype was cisAB01/O02.The serotype of samples 2 and 3 was A_(2)B,and the genotype was cisAB01/B101.The blood groups of samples 4 and 5 were AB and A,respectively.The genotyping results of ABO blood group of samples 1-3 were not consistent with the serological results.Conclusion The haplotype of cisAB01 and different ABO blood group alleles shows different phenotypes.Molecular biological detection technology is of great significance for the accurate identification of cisAB blood group,selection of appropriate blood for clinical transfusion and guarantee of blood transfusion safety.
