摘 要:目的建立复方木鸡颗粒中4种成分(金雀花碱、苦参碱、槐果碱和槲皮苷)含量的高效液相色谱法测定方法。方法采用Waters Xselect T3 C18(250 mm×4.6 mm,5μm)色谱柱;流动相为磷酸盐缓冲液(pH 6.8)-三乙胺(体积比70∶0.1)(A)-乙腈(B)-甲醇(C)进行梯度洗脱;流速1.0 mL·min-1,柱温30℃,检测波长220 nm。结果 4种成分的分离度良好,且在各自浓度范围内呈现出良好的线性关系。金雀花碱、苦参碱、槐果碱和槲皮苷在进样质量浓度分别为2.0~32.0、9.0~144.0、5.0~80.0和4.0~64.0 mg·L-1内呈良好线性关系,相关系数r≥0.999 0,平均回收率在95.3%~103.2%,RSD为3.8%~4.8%(n=9)。含量测定结果显示,3种生物碱和槲皮苷在不同批次中的含量比较稳定。结论该法简便、准确,分离效果好,可用于复方木鸡颗粒的多成分同时测定,为复方木鸡颗粒的质量控制提供方法参考。Objective To establish an HPLC method to simultaneously determine the contents of sophorine,matrine,sophocarpine and quercitrin in Fufang Muji granules,in order to provide basis for studying its quality standards.Methods An HPLC method was developed.Waters Xselect T3 C18 column(4.6 mm×250 mm,5 μm) was used and eluted with mobile phase of acetonitrile-methanol-phosphate buffer(pH6.8) at the flow rate of 1.0 mL·min-1.The wavelength was set at 220 nm.The column temperature was maintained at 30 ℃.Results The good linear relationships between the concentration and peak area were in the range of 2.0-32.0 mg·L-1 for cytosine,5.0-80.0 mg·L-1 for matrine,9.0-144.0 mg·L-1 for matrine and 4.0-64.0 mg·L-1 for quercitrin(r≥0.999 0),respectively.The average recoveries(n=9) were in the range of 95.3%-103.2% and RSD were in the range of 3.8%~4.8%.Conclusion The method is simple,feasible and reproducible and can be used for the quality control of Fufang Muji Granules.
