HPLC法同时测定大鼠血浆中白藜芦醇与木犀草素的含量

摘　　要：目的建立一种可同时测定大鼠血浆中白藜芦醇与木犀草素的HPLC法,并用于白藜芦醇-木犀草素双磷脂复合物在大鼠体内药动学行为的研究。方法采用Ecosil C18色谱柱(250 mm×4.6 mm,5μm)为固定相,流动相为甲醇-0.2%磷酸水溶液(体积比为66∶34),流速为1 mL·min^-1,检测波长为335 nm。通过乙酸乙酯液-液萃取法处理血浆,测定三只大鼠灌胃给予白藜芦醇-木犀草素双磷脂复合物后于不同时刻下血浆中白藜芦醇与木犀草素的质量浓度。测定结果以DAS2.0软件处理,得到白藜芦醇-木犀草素双磷脂复合物在大鼠体内的药动学参数。结果白藜芦醇与木犀草素分别在10.0~2000μg·L^-1和20.0~4000μg·L^-1内线性关系良好,两者定量下限分别为10.0μg·L^-1和20.0μg·L^-1,准确度(RE)均在-4.2%~9.7%之间,日内、日间精密度(RSD)均小于13%,提取回收率均在86.9%~94.3%之间。结论该方法可作为同时测定大鼠血浆中白藜芦醇与木犀草素质量浓度的方法,为探讨白藜芦醇-木犀草素双磷脂复合物在大鼠体内的药动学行为提供方法依据。Objective To establish an HPLC method for the simultaneous determination of resveratrol and luteolin in rat plasma,and to investigate the pharmacokinetics of resveratrol-luteolin phospholipid complex in rat after oral administration.Methods Three SD male rats were administrated with resveratrol-luteolin phospholipid complex,0.5 ml blood were collected at different time point respectively,and the plasma samples were extracted with ethyl acetate and analyzed by the HPLC method.Chromatographic separation of resveratrol and luteolin was performed on a Ecosil C18 column(250 mm×4.6 mm,5μm)with a mobile phase of methanol-0.2%phosphorous acid(V∶V=66∶34)at a flow-rate of 1.0 mL·min^-1,and UV detection was set at 335 nm.Pharmacokinetic parameters were calculated by DAS2.0 pharmacokinetic program.Results The linear calibration curve of resveratrol and luteolin was obtained respectively in the range of 10.0-2000μg·L^-1and 20.0-4000μg·L^-1,The lower limit of quantitation was 10.0μg·L^-1 and 20.0μg·L^-1;respectively;The accuracy(RE)were between-4.2%and 9.7%,the intra-day and inter-day precision(RSD)were less than 13%,and the recovery rate were within 86.9%-94.3%.Conclusion The method is simple,specific,sensitive and accurate.It is suitable for simultaneous determination of resveratrol and luteolin in rat plasma,and can be applied to investigate the pharmacokinetics of resveratrol-luteolin phospholipid complex in rats.
