摘 要:以细菌菌落数量、活性为指标,采用选择性培养法、革兰氏染色、芽孢染色、MTT法和平皿培养法研究了枸杞多糖对H22肝癌小鼠肠道大肠杆菌、双歧杆菌和乳酸杆菌的调节作用及其机制。结果表明:通过选择性培养后从菌落形态、革兰氏染色和芽孢染色成功鉴定培养的细菌分别是大肠杆菌、双歧杆菌和乳酸杆菌。肝癌小鼠肠道大肠杆菌显著增加,双歧杆菌和乳酸杆菌显著减少;而枸杞多糖对肝癌小鼠肠道大肠杆菌有抑制作用,对双歧杆菌和乳酸杆菌有促进作用。体外实验结果表明,枸杞多糖剂量在8~20 mg/mL时可以显著抑制大肠杆菌生长,而剂量在12~20 mg/mL时可促进乳酸杆菌生长,且都呈现剂量依赖性,但是对双歧杆菌无明显影响。由此可知,枸杞多糖对肝癌小鼠肠道大肠杆菌、双歧杆菌和乳酸杆菌有调节作用,部分机制可能与多糖直接抑制大肠杆菌、促进乳酸杆菌生长有关。The effects and mechanism of Lycium barbarum polysaccharide(LBP)on intestinal E.coli,Bifidobacteria and Lactobacillus in H22 hepatocellular carcinoma mice were investigated by using selective culture,Gram staining,spore staining,MTT method and plating method.The results showed that E.coli,Bifidobacteria and Lactobacillus were identified respectively by Gram staining and bacillus spore staining after selective culture.In H22-tumor bearing mice,intestinal E.coli increased significantly,while Bifidobacteria and Lactobacillus decreased significantly in quantity.LBP treatment could decrease E.coli while increase Bifidobacteria and Lactobacillus in quantity.The in vitro results showed that LBP at the doses of 8 mg/mL to 20 mg/mL could significantly inhibit the growth of E.coli.LBP at the doses of 12 mg/mL to 20 mg/mL could promote the growth of Lactobacillus directly,but showed little effect on Bifidobacteria.Taken together,LBP has regulatory effects on intestinal E.coli,Bifidobacteria and Lactobacillus in H22-tumor bearing mice,and part of the mechanism may be related to the direct inhibition of the polysaccharide on E.coli and the promotion of the growth of Lactobacillus.
