针灸联合柚皮素干预对腰椎间盘退变大鼠髓核NLRP3炎性小体的影响

摘　　要：目的探讨针灸联合柚皮素干预对腰椎间盘退变大鼠髓核NLRP3炎性小体的影响。方法将SD大鼠随机分成sham组、Model组、针灸组、柚皮素组、针灸+柚皮素组,每组10只。除sham组外,其他各组均采用腰椎纤维环损伤法构建腰椎间盘退变大鼠模型,造模成功后给予相应干预,每天1次,连续4周。HE染色观察椎间盘组织病理形态变化;免疫组织化学检测椎间盘组织中Ⅱ型胶原(CollagenⅡ)的表达;Tunel染色观察椎间盘髓核组织细胞凋亡水平;ELISA检测椎间盘髓核组织中炎症因子TNF-α、IL-1β和IL-18水平;qRT-PCR和Western blot检测椎间盘髓核组织中NLRP3、ASC和Caspase-1 mRNA和蛋白表达水平。结果与sham组比较,Model组大鼠椎间盘退变严重,椎间盘组织中CollagenⅡ表达显著降低(P<0.05),髓核细胞凋亡水平、髓核组织中NLRP3炎性小体相关基因mRNA及蛋白表达水平以及TNF-α、IL-1β和IL-18水平均显著上升(P<0.05)。与Model组比较,针灸组、柚皮素组和针灸+柚皮素组大鼠椎间盘退变程度均有所缓解,椎间盘组织中CollagenⅡ表达均显著升高(P<0.05),髓核细胞凋亡水平、髓核组织中NLRP3炎性小体相关基因mRNA及蛋白表达水平以及TNF-α、IL-1β和IL-18水平均显著下降(P<0.05)。其中针灸+柚皮素组改善效果最好。结论针灸联合柚皮素干预可减少髓核细胞凋亡和细胞外基质降解,抑制炎症反应,从而缓解大鼠腰椎间盘退变,其机制可能与抑制NLRP3炎性小体激活有关。Objective To investigate the effect of acupuncture combined with naringin on NLRP3 inflammasome of nucleus pulposus in rats with lumbar intervertebral disc degeneration.Methods SD rats were randomly divided into sham group,Model group,acupuncture group,naringin group,acupuncture+naringin group,with 10 rats in each group.Except for sham group,the lumbar intervertebral disc degeneration rat model was constructed by lumbar annulus fibroid injury method in other groups.After successful modeling,corresponding intervention was given,once a day for 4 weeks.HE staining was used to observe the pathological changes of intervertebral disc tissue.The expression of typeⅡcollagen(CollagenⅡ)in intervertebral disc tissue was detected by immunohistochemistry.Tunel staining was used to observe the apoptosis of intervertebral disc nucleus pulposus tissue.The expression of inflammatory factors TNF-α,IL-1βand IL-18 in intervertebral disc nucleus pulposus tissue were detected by ELISA.The mRNA and protein expression levels of NLRP3,ASC and Caspase-1 in intervertebral disc nucleus pulposus tissues were detected by qRT-PCR and Western blot.Results Compared with sham group,intervertebral disc degeneration was severe in Model group,CollagenⅡexpression in intervertebral disc tissue was significantly decreased(P<0.05).The apoptosis level of nucleus pulposus cells,the mRNA and protein expression levels of NLRP3 inflammasome related genes and the levels of TNF-α,IL-1βand IL-18 were significantly increased(P<0.05).Compared with Model group,the degree of intervertebral disc degeneration was alleviated in acupuncture group,naringin group and acupuncture+naringin group,CollagenⅡexpression in intervertebral disc tissue was significantly increased(P<0.05).The apoptosis level of nucleus pulposus cells,the mRNA and protein expression levels of NLRP3 inflammasome related genes and the levels of TNF-α,IL-1βand IL-18 were significantly decreased(P<0.05).Among them,acupuncture+naringin group had the best effect.Conclusion Acupuncture combin
