摘  要:目的研究渴络欣胶囊对2型糖尿病肾病(Diabetic nephropathy,DN)模型大鼠肾脏氧化应激和炎症反应的影响。方法通过注射链脲佐菌素(Streptozocin,STZ)和高脂肪、高糖饮食诱导DN模型大鼠,将模型大鼠随机分为DN组(模型组)、渴络欣组及厄贝沙坦组,每组8只。另选8只同龄大鼠作为正常组。正常组和模型组大鼠灌胃予生理盐水(10 mL·kg^(-1)·d^(-1)),渴络欣组大鼠灌胃予渴络欣胶囊制备的渴络欣混悬液(0.63 g·kg^(-1)·d^(-1)),厄贝沙坦组大鼠灌胃予厄贝沙坦混悬液(15.6 mg·kg^(-1)·d^(-1))。8周后,检测每组大鼠的空腹血糖(FBG)、尿微量白蛋白(MAU)、丙二醛(MDA)、超氧化物歧化酶(SOD)活性和炎症因子IL-1β、IL-6、IL-33、TNF-α、TGF-β的水平,并观察肾组织病理改变。结果模型组大鼠可见肾小球系膜增生、囊壁增生、肾小管扩张及上皮细胞气球样变等一系列病理改变,且肾脏纤维化严重,渴络欣治疗后上述肾脏病理现象减轻;与正常组相比,模型组大鼠SOD活性明显降低,MDA含量和IL-1β、IL-6水平显著升高(P<0.01);与模型组相比,渴络欣组大鼠SOD活性显著升高,MDA含量显著降低(P<0.01),IL-1β和TNF-α水平明显降低(P<0.05)。结论渴络欣胶囊能有效抑制糖尿病肾病模型大鼠的氧化应激和炎症水平,减轻肾脏病理学变化,延缓DN的病程进展。Objective To investigate the effect of Keluoxin capsule on oxidative stress and inflammatory response in the kidney of type 2 diabetic nephropathy(DN)model rats.Methods Rats induced DN models by injecting streptozocin(STZ)and a high-fat,high-glucose diet.Model rats were randomly divided into DN group(model group),Keluoxin group and Irbesartan group,with 8 rats in each group.8 normal rats of their same age were selected as normal group.Rats in the normal and model groups were gavaged to saline(10 mL·kg^(-1)·d^(-1)),rats in Keluoxin group were gavaged with a Keluoxin suspension prepared from Keluoxin capsules(0.63 g·kg^(-1)·d^(-1)),rats in Irbesartan group were gavaged with Irbesartan suspension(15.6 mg·kg^(-1)·d^(-1)).After 8 weeks,the levels of fasting blood glucose(FBG),urinary microalbumin(MAU),malledialdehyde(MDA),superoxide dismutase(SOD)activity and IL-1,IL-6,IL-33,TNF-α,TGF-βwere examined in each group,respectively.Results Pathological changes such as mesangial hyperplasia,cystic wall hyperplasia,renal tubular dilation and epithelial cell balloon-like changes were seen in the glomeruli of the model group,and the kidney fibrosis is serious.The above kidney pathology was reduced after treatment with Keluoxin.In the model group,the SOD activity was obviously reduced,and the MDA content、IL-1 and IL-6 levels were obviously increased compared with the normal group(P<0.01).In the Keluoxin group,the SOD activity was obviously increased,the MDA level was significantly decreased(P<0.01),and the levels of IL-βand TNF-αwere significantly reduced compared with the model group(P<0.05).Conclusion Keluoxin capsule can effectively inhibit oxidative stress and inflammation,reduce the kidney pathological changes of diabetic nephropathy model rats,and delay the progression of diabetic nephropathy.
