摘 要:目的鉴定与分析了虎杖中RGF多肽序列特征,阐明虎杖RGF多肽对植物根型的调控作用,为虎杖优形优质新品种奠定了理论研究基础。方法利用生物信息学的方法鉴定了虎杖RGF编码基因,并对其基序进行了比对、偏好性分析及系统进化分析。利用模式植物拟南芥根发育的体系,研究了虎杖PcRGF对根发育的影响。结果从虎杖基因组中鉴定出5个RGF基因(PcRGF1-5),系统进化分析表明PcRGF2-5与拟南芥AtRGF1、AtRGF4的亲缘关系较近,聚类为一族。PcRGF1与其他虎杖PcRGF较为分散,同拟南芥AtRGF9、AtRGF6的亲缘关系较近。与对照组相比,PcRGF2及PcRGF5影响根的重力反应,抑制了根的波浪(waving)表型。PcRGF4处理后主根长度相对于对照组显著增加,表明其促进根的生长。结论揭示了虎杖RGF家族的序列特征和系统进化关系,初步探究了虎杖PcRGF的生物学功能,发现虎杖PcRGF具备调控根发育的保守功能。Objective To identify RGF peptides in Polygonum cuspidatum and analyze its phylogenetic tree,and investigate the function of PcRGF in root development,which will provide the basis for molecular breeding of this medicinal plant.Methods By using bioinformatics approaches,RGF genes were identified in Polygonum cuspidatum.RGF motif,sequence logos and phylogenetic tree were analyzed.The PcRGF function was analyzed based on Arabidopsis root system.Results 5 RGF genes were identified in Polygonum cuspidatum.Phylogenetic analysis showed that PcRGF2-5 were clustered together and close to AtRGF1 and AtRGF4.However,PcRGF1 was not clustered with other PcRGF and closer to AtRGF9 and AtRGF6.PcRGF2 and PcRGF5 altered root gravity response and inhibited root waving phenotype.PcRGF4 promoted Arabidopsis root growth as compared with mock.Conclusion This paper reveals the sequences feature and phylogenetic relationship of PcRGF,investigates the function of PcRGF in root development,which the roots of Polygonum cuspidatum are designed as medical use.
