摘  要:目的考察蜜桑白皮的体内降脂减肥作用。方法建立小鼠高脂肥胖模型,随机分为正常对照组、肥胖模型组、蜜桑白皮灌胃组、阳性对照组,记录小鼠体重,测定肾周脂肪指数和附睾脂肪指数,及血清中总胆固醇、甘油三酯、高密度脂蛋白胆固醇和低密度脂蛋白胆固醇水平。此外,通过Autodock模拟蜜桑白皮中主要成分与脂质代谢关键酶——羟甲基戊二酰辅酶A还原酶(HMGR)及羧酸酯酶I(hCES1A)的相互作用,预测其调节血脂作用机制。结果蜜桑白皮组可显著减缓小鼠体重增加,并有效降低LDL-C水平,其效果优于阳性奥利司他药物组。经Autodock分子对接发现,主要成分桑色素及桑根酮C可分别通过氢键与HMGR的催化活性中心Asp-767、及hCES1A关键氨基酸Cys-389产生较强的相互作用。结论蜜桑白皮具有良好的减肥降脂作用。Objective To investigate the in vivo fat loss effects of honey-processed Cortex Mori.Methods A mouse high-fat obesity model was established and randomly divided into normal control group,obesity model group,honeyprocessed Cortex Mori gavage group and positive control group.The body weight of mice was recorded,perirenal adiposity index and epididymal adiposity index,and serum levels of total cholesterol,triglyceride,HDL cholesterol and LDL cholesterol were measured.In addition,the interaction of the main components of honey-processed Cortex Mori with 3-hydroxy-3-methylglutaryl coenzyme A reductase(HMGR)and carboxylesterase I(hCES1 A),the key enzymes of lipid metabolism,was simulated by Autodock to predict the mechanism of its lipid-regulating effects.Results The honey-processed Cortex Mori group could significantly slow down the weight gain of mice and effectively reduce the level of LDL-C,which was better than the positive orlistat group.Docking simulations demonstrated that Morin and Sanggenone C could interact strongly with HMGR and hCES1 A through hydrogen bonds,respectively.Conclusion The honey-processed Cortex Mori had a potent effect on obesity alleviation.
