摘 要:目的观察消癌解毒方对H22荷瘤小鼠热休克蛋白表达水平的影响,探求消癌解毒方治疗肝癌的潜在分子机制。方法设置模型组及消癌解毒方高、中、低剂量组和顺铂阳性对照组共5组,药物干预治疗后,观察各组瘤体比,病理染色观察肿瘤细胞增殖情况,RT-PCR法和Western blot法检测肿瘤组织中HSP27、HSP60、HSP70的基因和蛋白表达水平。结果治疗11 d后,与模型组相比,治疗组瘤重均出现不同程度下降(P<0.01),且以高浓度组差异最为明显;病理染色显示,与模型组比较,治疗组肿瘤细胞的核分裂现象减少;RT-PCR和Western blot检测结果提示,与模型组相比,治疗组中HSP27基因和蛋白表达均出现不同程度的下调及HSP60、HSP70基因和蛋白表达的上调,其中以消癌解毒方高剂量组效果最佳。结论消癌解毒方可显著抑制H22型肝癌细胞的增殖情况,这一抗肿瘤作用可能与下调HSP27表达,上调HSP60、HSP70表达相关,且抑瘤作用呈现药物浓度依赖性,即药物浓度越高,抑瘤作用越强。Objective To observe the effect of Xiaoai Jiedu Recipe on the expression of heat shock protein(HSP) in H22 tumor bearing mice,and explore the potential molecular mechanism of Xiaoai Jiedu Recipe in the treatment of liver cancer. Methods The mice were divided into five groups,including model group,cisplatin positive control group,Xiaoai Jiedu Recipe groups with high-,medium-and low-dose.After drug intervention,the tumor/body ratio of each group was observed,and the proliferation of tumor cells was observed by pathological staining.The mRNA and protein expression levels of HSP27,HSP60 and HSP70 were detected by RT-PCR and Western blot. Results After treatment for 11 days,compared with the model group,the tumor weight in the treatment groups was decreased in different degrees(P<0.01),and the effect was most significant in the high-dose group.Pathological staining showed that,compared with the model group,the mitosis of tumor cells was decreased in the treatment groups.The results of RT-PCR and Western blot suggested that compared with the model group,HSP27 gene and protein expressions were down-regulated in the treatment groups with different degrees,and the gene and protein expressions of HSP60 and HSP70 were up-regulated,and the effect was most significant in the Xiaoai Jiedu Recipe group with high-dose. Conclusion Xiaoai Jiedu Recipe can significantly inhibit the proliferation of H22 hepatoma cells,and its anti-tumor effect may be related to down-regulation of HSP27 expression and up-regulation of HSP60 and HSP70 expressions.The anti-tumor effect shows in a dose-dependent manner,that means the higher the drug concentration,the greater the anti-tumor effect.
