摘  要:目的:观察四氯化碳(CCl_(4))及二甲基亚硝胺(DMN)诱导小鼠肝纤维化模型肝脏淋巴细胞亚群变化特点,并进一步观察中药扶正化瘀方对淋巴细胞亚群变化的调节作用。方法:分别采用10%CCl_(4)及0.5%DMN腹腔注射制备小鼠肝纤维化模型,观察小鼠肝脏大体观及脏器指数,检测小鼠血清ALT、AST表达变化,HE和天狼猩红染色观察肝组织炎症及纤维化程度,流式细胞术观察小鼠肝脏淋巴细胞亚群比例和数量变化。结果:与正常组比较,两种模型组小鼠肝脏表面粗糙,质地变硬,粟粒样表现明显,肝体比下降,脾体比升高,血清ALT、AST表达增加,肝组织病理染色显示明显的炎症和胶原沉积,肝脏NK细胞、NKT细胞、CD4^(+)T细胞比例和数量下降,CD8^(+)T细胞和B细胞比例和数量升高(P<0.05,P<0.01);与模型组比较,药物组小鼠肝脏大体观明显改善,肝体比增加,脾体比下降,血清ALT、AST表达下降,肝组织炎症和胶原沉积明显减轻,肝脏NK细胞、NKT细胞、CD4^(+)T细胞比例和数量升高,CD8^(+)T细胞和B细胞比例和数量下降(P<0.05,P<0.01)。结论:CCl_(4)和DMN诱导肝纤维化模型小鼠肝脏淋巴细胞亚群免疫紊乱,中药扶正化瘀方可调节肝脏免疫微环境,纠正免疫紊态,发挥抗肝纤维化作用。ABSTRCT Objective:To observe the characteristics of changes of intrahepatic lymphocyte subsets in mice models with hepatic fibrosis induced by carbon tetrachloride(CCl_(4))and dimethylnitrosamine(DMN),and to further observe the regulatory effect of traditional Chinese medicine Fuzheng Huayu Formula on lymphocyte subsets changes.Methods:The mouse models with hepatic fibrosis were established by intraperitoneal injection of 10%CCl_(4)and 0.5%DMN respectively.The general view and organ indexs of the mouse liver were observed,the changes of serum ALT and AST levels were detected,HE and Sirius Red staining were used to evaluate the degrees of liver inflammation and fibrosis,and flow cytometry was used to observe the changes in the proportion and number of mouse liver lymphocyte subsets.Results:Compared with the normal group,the livers of mice in the two model groups were rough on the surface,hard in the texture and the miliary appearance was obvious.The liver-to-body ratio was decreased,while the spleen-to-body ratio was increased.Serum ALT and AST expressions were increased,and the liver histopathological staining showed obvious inflammation and collagen deposition.The proportions and numbers of all intrahepatic NK cells,NKT cells,and CD4^(+)T cells were decreased,and the proportions and numbers of CD8^(+)T cells and B cells were increased(P<0.05,P<0.01).Compared with the model group,the general view of livers of the drug groups was significantly improved.The liver-to-body ratio was increased,while the spleen-to-body ratio was decreased(P<0.05,P<0.01).Serum ALT and AST expressions were decreased.Inflammation and collagen deposition in liver tissue were reduced significantly.The proportions and numbers of all intrahepatic NK cells,NKT cells,and CD4^(+)T cells were increased,while the proportions and numbers of CD8+T cells and B cells were decreased(P<0.05,P<0.01).Conclusion:CCl_(4)and DMN can induce immune disorders of lymphocyte subsets in hepatic fibrosis model mice,and the Chinese medicine Fuzheng Huayu Formula can
