摘 要:为探讨NK细胞和绒毛外滋养(extravillous trophoblast,EVT)细胞与子宫血管重塑间的关系,采用免疫组化连续切片和动物实验,检测NK细胞及EVT细胞在子宫动脉的浸润情况,并观察不同妊娠阶段子宫动脉的变化。结果显示,孕5~8周的人子宫血管鲜有重塑;孕9~13周的人子宫动脉显示出中等程度的重塑,血管周围有大量NK细胞和少许EVT细胞聚集;孕14~20周以及孕21~28周,子宫血管重塑明显,周围NK细胞较少而EVT细胞明显增多。采用腹腔注射抗asialo GM1血清去除小鼠体内NK细胞,导致小鼠子宫动脉重塑严重受阻,小鼠出现子痫前期(preeclampsia,PE)样症状。提示NK细胞可能通过与EVT细胞协作参与妊娠早期子宫血管重塑,子宫动脉重塑程度与孕周相关。To investigate the relationship among NK cells and extravillous trophoblast(EVT)cells and uterine vascular remodeling,the immunohistochemical serial sections and animal experiments were used to detect the infiltration of NK cells and EVT cells in the uterine artery and observe changes in uterine arteries at different stages of pregnancy.The results showed that few uterine vessels remodeled during 5 to 8 weeks gestation.Vessels of 9 to 13 weeks of gestation demonstrated moderate remodeling with abundant NK cells and a few EVT cells surrounding the vessel.Vessels of 14 to 20 and 21 to 28 weeks of gestation displayed substantial remodeled changes with a large number of EVT cells and decrease NK cells around the vessel.Depletion of NK cells using anti-asialo GM1 serum led to impaired uterine vessel remodeling and preeclampsia(PE)symptoms in pregnant mice.The results demonstrate that NK cells may play an important role in uterine vascular remodeling in collaboration with EVT cells,and the degree of uterine artery remodeling is associated with gestational age.
