摘  要:目的:探究南蛇藤提取物通过抑制磷酯酰肌醇-3-激酶(PI3K)/蛋白激酶B(AKT)信号通路对胃癌细胞转移和侵袭的影响。方法:取对数生长期的人胃癌细胞系AGS细胞,设为对照组(vector)、阳性药物组(cisplatin)、低剂量组(low)、中剂量组(medium)和高剂量组(high),vector组以培养液培养,cisplatin组以顺铂处理,low、medium和high组以不同剂量(10、50、100μg/mL)的南蛇藤提取物处理,分别检测各组细胞增殖、迁移、侵袭和凋亡水平,并检测细胞内上皮间充质转化标记物(E-cadherin、N-cadherin、Vimentin)和PI3K/AKT信号通路的蛋白表达。结果:与vector组相比,cisplatin、low、medium和high组细胞增殖率均显著降低,且呈时间和剂量依赖性(P<0.05)。与vector组相比,cisplatin、low、medium和high组细胞迁移和侵袭能力均被显著抑制,且随着南蛇藤提取物剂量的增加,迁移、侵袭抑制效果更明显(P<0.05)。与vector组相比,cisplatin、low、medium和high组细胞凋亡率均增加,且随着南蛇藤提取物剂量的增加,细胞凋亡率随之上升(P<0.05)。与vector组相比,cisplatin、low、medium和high组E-cadherin的表达显著上调,N-cadherin和Vimentin的表达显著下调,且呈剂量依赖性。与vector组相比,cisplatin、low、medium和high组PI3K、AKT的蛋白表达被显著下调,且随着南蛇藤提取物剂量的增加,下调幅度更大。结论:南蛇藤提取物通过抑制PI3K/AKT信号通路的活化,抑制胃癌细胞体外增殖、迁移、侵袭和上皮间充质转化,诱导细胞凋亡,发挥显著的抗肿瘤效应。Objective:To explore the effect of celastrus orbiculatus extracts on the metastasis and invasion for gastric cancer cells by inhibiting the phosphatidylinositol-3-kinase(PI3 K)/protein kinase B(AKT)signaling pathway.Methods:AGS cells of human gastric cancer cell line in logarithmic phase were collected as a controlled group(vector),a positive drug group(cisplatin),a low-dose group(low),a medium-dose group(medium)and a high-dose group(high).The vector group was cultured with culture medium,the cisplatin group was treated with cisplatin,and the low,medium and high groups were treated with different doses(10,50,100μg/mL)of celastrus orbiculatus extracts.The proliferation,migration,invasion and apoptosis of each group were detected respectively,and the protein expressions of intracellular epithelial-mesenchymal transition markers(E-cadherin,N-cadherin,Vimentin)and PI3 K/AKT signaling pathway were detected.Results:Compared with the vector group,the cell proliferation rates of the cisplatin,low,medium and high groups was significantly reduced in a time-and dose-dependent manner(P<0.05).The cell migration and invasion of the cisplatin,low,medium and high groups were all significantly inhibited compared to the vector group,and with the increase in the dose of celastrus orbiculatus extracts,the inhibition effect on migration and invasion was more obvious(P<0.05).Compared with the vector group,the apoptosis rate of the cisplatin,low,medium and high groups was all increased,and the apoptosis rate was risen with the increase of dose of celastrus orbiculatus extracts(P<0.05).The expression of E-cadherin in the cisplatin,low,medium and high groups was significantly up-regulated while the expressions of N-cadherin and Vimentin were significantly down-regulated compared to the vector group in a dose-dependent manner.The protein expressions of PI3 K and AKT were significantly down-regulated in the cisplatin,low,medium and high groups compared to the vector group,and with the increase in the dose of celastrus orbiculatus extracts
