养心活血汤加减对扩张型心肌病小鼠心肌Bcl-2及Bax蛋白表达的影响  被引量:1

摘  要:目的:观察雷忠义教授自拟养心活血汤加减对阿霉素诱导的扩张型心肌病(Dilated cardiomypathy,DCM)小鼠心肌Bcl-2和Bax蛋白表达的影响,探讨养心活血汤加减治疗DCM可能的分子机制。方法:将30只小鼠随机分为正常对照组(对照组,A组)、阿霉素诱导的DCM模型组(模型组,B组)、养心活血汤加减干预组(药物干预组,C组)。分别检测小鼠血压、心率,应用心动超声、HE染色、TUNEL检测、Western blot检测小鼠心脏功能、心肌组织细胞病理形态、心肌细胞凋亡率、Bax及Bcl-2蛋白表达水平等。结果:1.与对照组相比,模型组、小鼠LVEDD、LVESD均升高(P<0.01),FS、EF降低(P<0.01);与模型组相比,药物干预组小鼠LVEDD、LVESD均降低(P<0.01),FS、EF升高(P<0.01);2.与对照组相比,模型组收缩压、舒张压明显降低(P<0.01);与模型组相比,药物干预组收缩压、舒张压无明显统计学差异(P=0.944);3.与对照组相比,TUNEL检测模型组小鼠心肌凋亡细胞数明显增多(P<0.01),与模型组相比,药物干预组凋亡细胞明显减少(P<0.01),HE染色见镜下模型组小鼠心肌细胞损伤、心肌纤维化、炎细胞浸润明显,而药物干预组小鼠心肌细胞明显改善。4.与对照组相比,模型组小鼠Bcl-2的蛋白表达量显著降低(P<0.01),而Bax蛋白表达量明显增加(P<0.01);与模型组相比,药物干预组小鼠心肌的Bcl-2蛋白表达量增加(P<0.01),Bax蛋白表达量明显下降(P<0.01)。结论:养心活血汤加减通过调控Bcl-2及Bax蛋白表达来保护DCM小鼠心肌细胞,在改善DCM小鼠的心功能同时而不影响血压。Objective:To observe the effect of Modified Self-made Yangxin Huoxue Decoction that of Prof.LEI Zhongyi’s experience in the expression of Bcl-2 and Bax protein in the myocardium of dilated cardio-myopathy model mice,and to explore the possible molecular mechanism of Modified Yangxin Huoxue Decoction in the treatment of DCM.Methods:Thirty mice were randomly divided into a normal controlled group,a model group,and a drug intervention group.The blood pressure and heart rate of the mice were detected respectively,and cardiac function,myocardial tissue cell pathological morphology,myocardial cell apoptosis rate,Bax and Bcl-2 protein expression levels were detected by echocardio-graphy,HE staining,TUNEL detection and Western blot.Results:1.Compared with the controlled group,the LVEDD and LVESD of the model group and mice were increased(P<0.01),while the FS and EF were decreased(P<0.01).Compared with the model group,the LVEDD and LVESD in the drug intervention group were decreased(P<0.01),while the FS and EF were increased(P<0.01);2.Compared with the controlled group,the systolic blood pressure and diastolic blood pressure in the model group were significantly lower(P<0.01);Compared with the model group,there was no significant difference in systolic and diastolic blood pressure in the drug intervention group(P=0.944);3.Compared with the controlled group,the number of myocardial apoptotic cells in the TUNEL detection model group was significantly increased(P<0.01).Compared with the model group,the apoptotic cells in the drug intervention group were significantly reduced(P<0.01).HE staining showed that the myocardial cell injury,myocardial fibrosis and inflammatory cell infiltration were obvious in the model group mice,while the myocardial cells in the drug intervention group were significantly improved.4.Compared with the controlled group,the protein expression of Bcl-2 in the model group mice was significantly decreased(P<0.01),while the protein expression of Bax was significantly increased(P<0.01).Compared with the
