摘  要:目的:从NOD样受体蛋白3(NLRP3)信号通路,探究冠通方改善大鼠血管内膜损伤后再狭窄的分子机制。方法:将大鼠随机分为假手术组、模型组、冠通方组、NLRP3激活剂组、冠通方+激活剂组,每组12只;用颈动脉球囊损伤术复制血管内膜损伤后再狭窄模型,各组大鼠经药物干预处理4周后,ELISA法检测大鼠血管白细胞介素-1β(IL-1β)、肿瘤坏死因子-α(TNF-α)、单核细胞趋化蛋白-1(MCP-1)水平;HE染色观察颈动脉血管组织病理变化;增殖细胞核抗原(PCNA)免疫组化染色法检测血管平滑肌细胞增殖状况;RT-PCR法检测血管组织NLRP3mRNA、IL-1βmRNA表达;Western blot法检测血管组织NLRP3、凋亡相关斑点样蛋白(ASC)、IL-1β、半胱氨酸天冬氨酸蛋白水解酶1(Caspase-1)蛋白表达。结果:与假手术组相比,模型组大鼠颈总动脉血管组织内膜增生及管腔狭窄等病理损伤加重,内膜增生程度、炎症因子水平、血管平滑肌细胞增殖活性及NLRP3炎性小体信号通路活性升高(P<0.05)。冠通方干预治疗后,大鼠血管内膜增生、管腔狭窄等病理损伤减轻,内膜增生程度、炎症因子水平、血管平滑肌细胞增殖活性及NLRP3炎性小体信号通路活性均降低(P<0.05)。NLRP3激活剂可减弱冠通方抑制NLRP3炎性小体信号活性、降低炎症、改善内膜增生、抗血管狭窄等作用(P<0.05)。结论:冠通方可能通过抑制NLRP3炎症小体通路活化,降低血管炎症反应,发挥抗血管再狭窄作用。Objective:From the NOD-like receptor protein 3(NLRP3)signaling pathway,to explore the molecular mechanism of Guantong Recipe in improving restenosis after intimal vascular injury in rats.Methods:The rats were randomly divided into a sham operation group,a model group,a Guantong Recipe group,a NLRP3 activator group,a Guantong Recpe+activator group,with 12 rats in each group,carotid artery balloon injury was used to replicate the restenosis model after vascular intimal injury,after the rats in each group were treated with drug intervention for 4 weeks,ELISA method was used to detect the levels of vascular interleukin-1β(IL-1β),tumor necrosis factor-α(TNF-α)and monocyte chemoattractant protein-1(MCP-1).HE staining was used to observe the pathological changes of carotid vascular tissue,proliferating cell nuclear antigen(PCNA)immunohistochemical staining method was used to detect the proliferation of vascular smooth muscle cells,RT-PCR method was used to detect the expression of NLRP3 mRNA and IL-1βmRNA in vascular tissues,Western blot was used to detect the expression of NLRP3,apoptosis-related dot-like protein(ASC),IL-1β,and Caspase-1 protein in vascular tissues.Results:Compared with the sham operation group,the intimal hyperplasia and lumen stenosis of the common carotid artery in the model group were worsened,the degree of intimal hyperplasia,the level of inflammatory factors,the proliferation activity of vascular smooth muscle cells and the activity of NLRP3 inflammasome signaling pathway were increased(P<0.05).After Guantong Recipe intervention treatment,the pathological damages such as vascular intimal hyperplasia and lumen stenosis were alleviated,the degree of intimal hyperplasia,the level of inflammatory factors,the proliferation activity of vascular smooth muscle cells and the activity of NLRP3 inflammasome signaling pathway were all reduced(P<0.05).NLRP3 activator could attenuate the effects of Guantong Recipe on inhibiting NLRP3 inflammatory body signal activity,reducing inflammation,improving intim
