摘 要:【目的】探讨复方七芍降压片(三七、白芍、三麻等组成)对自发性高血压大鼠(SHR)肾动脉重塑的干预作用及机制。【方法】将SHR随机分为模型对照组、厄贝沙坦片组、复方七芍降压片组,每组10只,另设10只WKY大鼠为正常对照组。厄贝沙坦片组给予厄贝沙坦片混悬液27 mg·kg^(-1)·d^(-1)灌胃,复方七芍降压片组给予复方七芍降压片混悬液496.8 mg·kg^(-1)·d^(-1)灌胃,正常对照组和模型对照组给予等体积蒸馏水10 m L·kg^(-1)灌胃。每天给药1次,连续给药6周。给药结束后,苏木素-伊红(HE)染色法观察大鼠肾动脉组织病理变化,并测量管壁厚度、中膜厚度及相同位置的管腔直径,计算中膜厚度/管腔直径比值;免疫组织化学法检测肾动脉中Ⅰ型胶原(COⅠ)、Ⅲ型胶原(COⅢ)蛋白表达情况。【结果】与正常对照组比较,模型对照组大鼠肾动脉管壁厚度、中膜厚度/管腔直径比值增高(P<0.01),肾动脉组织COⅠ、COⅢ表达水平均明显升高(P<0.01);与模型对照组比较,复方七芍降压片组和厄贝沙坦片组大鼠肾动脉管壁厚度、中膜厚度/管腔直径比值降低(P<0.05),肾动脉组织COⅠ、COⅢ表达水平均明显降低(P<0.05或P<0.01),2个给药组间差异无统计学意义(P>0.05)。【结论】复方七芍降压片可改善SHR肾动脉重塑,其机制与抑制平滑肌细胞增殖、迁移及COⅠ、COⅢ胶原蛋白沉积有关。Objective To explore the intervention effect and mechanism of Compound Qishao Antihypertensive Tablets for renal artery remodeling in spontaneously hypertensive rats(SHR).Methods The SHR were randomly divided into model control group,Irbesartan Tablets group and Compound Qishao Antihypertensive Tablets group,with 10 rats in each group,and 10 WKY rats were set up as normal control group.The Irbesartan Tablets group was given Irbesartan Tablets suspension at 27 mg·kg^(-1)·d^(-1)by gavage,the Compound Qishao Antihypertensive Tablets group was given Compound Qishao Antihypertensive Tablets suspension at 496.8 mg·kg^(-1)·d^(-1)by gavage,and the normal control group and the model control group were given an equal volume of distilled water at 10 mL·kg^(-1)by gavage.The administration was performed once daily for 6 continuous weeks.After the administration,the pathological changes in the renal arteries of rats were observed by hematoxylin-eosin(HE)staining,and the thickness of the vessel wall,the thickness of the mid-membrane and the diameter of lumen at the same location were measured,and the ratio of mid-membrane thickness/lumen diameter was calculated;the protein expression of collagen typeⅠ(COⅠ)and collagen typeⅢ(COⅢ)in the renal arteries was measured by immunohistochemistry.Results Compared with the normal control group,the renal artery wall thickness and intima-media thickness/lumen diameter ratio were increased in the model control group(P<0.01),and the expression levels of COⅠand COⅢin the renal artery tissues were significantly increased(P<0.01);compared with the model control group,the renal artery wall thickness and intima-media thickness/lumen diameter ratio were decreased in the Compound Qishao Antihypertensive Tablets group and Ivbesartan Tablets group(P<0.05),the expression levels of COⅠand COⅢin renal artery tissues were significantly reduced(P<0.05 or P<0.01),and there was no statistically significant difference between the two medication groups(P>0.05).Conclusion Compound Qishao Ant
