摘 要:疫苗接种是防治传染性疾病的一种有效的防治策略。安全、有效和低成本的疫苗是如今疫苗研究的追求目标。生物信息学和高通量测序的方法为疫苗研究带来了新的思路,通过生物信息学分析工具进行表位预测,分析出可以诱导有效的细胞免疫和体液免疫反应的优势表位参与疫苗构建,再综合其他方面的生物信息学技术评估最终构成新的多表位疫苗。本文主要从抗原选取、计算机表位预测、多表位的连接、蛋白三维构建及评估、分子对接、分子动力学模拟等方面对当前最主要的多表位疫苗研究策略进行综述,同时总结了多表位疫苗目前的研究情形和未来的发展。该策略已经应用于新冠病毒、疟疾及利什曼原虫等病原体的多表位疫苗研究之中。Vaccination is an effective control strategy against infectious diseases.A safe,effective,and cost-efficient vaccine is the aim of vaccine research.Bioinformatics and high-throughput sequencing approaches have brought new ideas to vaccine research.Through the prediction of epitopes by bioinformatics tools,we can analyze the dominant epitopes that can induce effective cellular and humoral immune responses for the construction of vaccines,and finally integrate other bioinformatics to evaluate to new multi-epitope vaccines.In this paper,we reviewed the most current multi-epitope vaccine research strategies including antigen selection,epitope prediction in silico,linkage of multiple epitopes,tertiary structure construction and evaluation,molecular docking,molecular dynamics simulation,and also summarize the current research situation and future development of multi-epitope vaccines.The strategy has been applied to the study of multi-epitope vaccines for pathogens such as 2019 novel coronavirus,malaria,and Leishmania.
