银翘散联合克拉霉素对蜃楼弗朗西斯菌的抑制作用研究

摘　　要：目的探究银翘散、克拉霉素对蜃楼弗朗西斯菌的体外抑制作用。方法选取蜃楼弗朗西斯菌ATCC25015为实验菌株,采用微量肉汤稀释法测定银翘散和克拉霉素的最低抑菌浓度(MIC);采用微量棋盘稀释法进行银翘散和克拉霉素的联合药敏试验;利用紫外分光光度法及RT-PCR测定银翘散和克拉霉素单用或联用时对蜃楼弗朗西斯菌生物膜形成的影响及greA基因mRNA表达的变化。结果银翘散、克拉霉素单药对蜃楼弗朗西斯菌MIC分别为6.40、0.32 mg/mL,二者联合用药时分级抑菌浓度指数(FIC)为0.75,提示两药配伍具有相加作用。与空白对照组比较,银翘散(1.6 mg/mL)、克拉霉素(0.08 mg/mL)可抑制蜃楼弗朗西斯菌生物膜的形成能力(P<0.05),而两药联合使用可增强抑制生物膜形成的能力,且较相同浓度单药组MIC浓度下降。与空白对照组比较,RT-PCR表明一定浓度的银翘散、克拉霉素单药或联用后菌株greA mRNA表达水平均呈现下调(P<0.05),但联合用药组较相同浓度的克拉霉素单药组下调更显著(P<0.05)。结论银翘散与克拉霉素联用时对蜃楼弗朗西斯菌的体外抑菌呈相加作用,同时可以增强对其生物膜形成及greA mRNA表达的抑制作用。Objective To explore the in vitro inhibitory effects of Yinqiao powder(Y)and clarithromycin(K)on Francisella philomiragia.MethodsThe Francisella philomiragia ATCC25015 was cultured as the experimental strain.The minimum inhibitory concentrations(MICs)of Yinqiao powder and clarithromycin were determined by the broth dilution method,and the fractional inhibitory concentration(FIC)index of part of them was calculated to determine the antibacterial effect of combined drugs.Ultraviolet spectrophotometry was used to determine the effect of Yinqiao powder and clarithromycin on the formation of Francisella philomiragia biofilms.The change of expression of greA was detected by Real-time fluorescence quantitative RT-PCR.ResultsThe MICs of Yinqiao powder and clarithromycin were 6.40 and 0.32 mg/mL,respectively.After the combination of two-drugs,FIC was 0.75,indicating that these two drugs had an additive effect.Compared with the control group,Yinqiao powder(1.6 mg/mL)and clarithromycin(0.08 mg/mL)could inhibit the formation of Francisella philomiragia biofilms(P<0.05)and the combination of both drugs archiving the same inhibitory effect with lower drug concentrations(P<0.05).In the meanwhile,the RT-PCR results showed that the expression level of greA mRNA was downregulated by Yinqiao powder and clarithromycin(P<0.05),and the combination of both drugs at the same concentration had a higher down regulation effect(P<0.05).ConclusionYinqiao powder had an additive effect with clarithromycin on Francisella philomiragia in vitro,and at the same time it could enhance the inhibitory effect on the formation of biofilm and the expression of greA mRNA.
