福建武夷山地区土壤可培养放线菌多样性分析

摘  要:采用培养法连续两年对福建武夷山地区24个定位观测点土壤放线菌进行分离培养和纯化,并应用16S rRNA基因进行物种鉴定及其多样性分析.结果表明:(1)分离获得的109株放线菌可包含了5个目、6个科、10个属,10个属分别为链霉菌属(Streptomyces)、节杆菌属(Arthrobacter)、假节杆菌属(Pseudarthrobacter)、类节杆菌属(Paenarthrobacter)、北里孢菌属(Kitsatospora)、小单孢菌属(Micromonospora)、中国单胞菌属(Sinomonas)、拟诺卡氏菌属(Nocardiopsis)、类诺卡氏菌属(Nocardiodies)和野野村菌属(Nonomuraea).(2)分离获得链霉菌属菌株最多(79株),有42个种,是武夷山地区土壤可培养放线菌的优势菌属;其次是北里孢菌属(17株),有5个种.慧苑坑土壤分离获得的放线菌物种最多(9种),而麻粟样地海拔1280 m处只分离到1种.(3)黄溪洲海拔1200 m和鬼洞处土壤中放线菌群落的Simpson和Shannon指数均为最高,茶洞的Simpson和Shannon指数均最低.Soil actinomycetes were isolated,cultured and purified from 24 sites in Wuyishan area of Fujian Province for two consecutive years,species identification and diversity analysis were performed using 16 S rRNA gene.The results showed(1)A total of 109 strains of actinomycetes were obtained from the soils in Wuyishan area of Fujian Province,which could be classified into 5 orders,6 families and 10 genera respectively,including the genera of Streptomyces,Arthrobacter,Pseudarthrobacter,Paenarthrobacter,Kitsatospora,Micromonospora,Sinomonas,Nocardiopsis,Nocardiodies and Nonomuraea;(2)Streptomyces was the dominant taxon at the genus level of cultivable actinomycetes in the Wuyishan area of Fujian Province,and there were 42 species consisting of 79 strains,Kitsatospora was the second dominant taxon of the actinomycetes isolated,including 5 species;Huiyankeng had the most abundant species of actinomycetes(9 species)in the soils among the 24 sites,while only one species of actinomycetes was isolated from the site of Masu at an altitude of 1280 m;(3)The actinomycete community diversity index showed that the indexes of Simpson and Shannon at the site of Huangxizhou at an altitude of 1200 m and Guidong were the highest,while those of Chadong were the lowest.
