摘 要:目的观察高度纯化的副溶血弧菌(Vibrio parahaemolyticus)噬菌体Qdvp001和Vpp1对外周血单核细胞(peripheral blood mononuclear cell,PBMCs)表达IL-1β、IL-6、CD14免疫基因的调控。方法①对噬菌体Qdvp001和Vpp1进行纯化;②分别用噬菌体低滴度组、中滴度组、高滴度组,Vp17802组和对照组作用于PBMCs后,用MTT比色法检测细胞增殖活性;③选取噬菌体中滴度组、Vp17802组、噬菌体+Vp17802组和对照组作用于PBMCs后,RT-PCR检测IL-1β、IL-6、CD14基因mRNA的变化。结果分别经两次纯化后,Qdvp001内毒素含量降低了644倍,Vpp1内毒素含量降低了652倍。噬菌体低、中、高滴度组作用于PBMCs一段时间后,细胞的增殖活性与对照组相比差异无统计学意义(P>0.05)。与对照组相比两种噬菌体对IL-1β和IL-6以及CD14的表达差异均无统计学意义(P>0.05)。与细菌组相比,噬菌体Qdvp001+Vp17802组先促进后抑制IL-1β的表达(P<0.05),一直抑制IL-6和CD14的表达(P<0.05);噬菌体Vpp1+Vp17802组一直抑制IL-1β和IL-6以及CD14的表达(P<0.05)。结论高度纯化的副溶血弧菌噬菌体对PBMCs增殖活性无明显影响且能够调节细菌引起的免疫应答。Objective To observe the regulation of highly purified Vibrio parahaemolyticus bacteriophage Qdvp001 and Vpp1 on inducing peripheral blood mononuclear cell(PBMCs)to express immune genes for IL-1β,IL-6,and CD14.Methods①Phage Qdvp001 and Vpp1 were purified;②After using the tested phage groups with low,median and high titer,Vp17802 and control group on PBMCs,the cell proliferation activity was detected by MTT colorimetry;③After selecting phage with median titer,Vp17802,phage+Vp17802 and control group on PBMCs,RT-PCR was used to detect the changes of mRNA for IL-1β,IL-6 and CD14 genes.Results Performed two purifications,the content of endotoxin in Qdvp001 and Vpp1 decreased 644 times and 652 times,respectively.There was no a statistical significance in cell growth treated with the low,median and high titer phage(P>0.05).Compared with the control group,the expression of IL-1β,IL-6 and CD14 was no a statistical significance(P>0.05)by the two phages.Compared with the Vp17802 group,the Qdvp001+Vp17802 was promoted first and then inhibited the expression of IL-1β(P<0.05),and it has consistantly inhibited the expression of IL-6 and CD14(P<0.05);the Vpp1+Vp17802 has consistantly inhibited the expression of IL-1β,IL-6 and CD14(P<0.05).Conclusion The highly purified Vibrio parahaemolyticus phage has no a significant effect on the proliferation activity of PBMCs and could regulate the immune response caused by bacteria.
