摘 要:目的探讨角叉菜胶构建大鼠慢性非细菌性前列腺炎(CNP)模型的影响因素。方法选用成年雄性大鼠注射角叉菜胶造模,观察鼠种(SD大鼠、Wistar大鼠)、注射部位(精囊、前列腺腹侧叶)、注射浓度(1%、3%)对建模的影响,根据前列腺组织病理学指标HE染色结果判定建模效果。结果(1)SD雄性大鼠前列腺组织病理HE染色结果较Wistar雄性大鼠明显(P<0.05);(2)Wistar雄性大鼠正常组部分出现自发性炎症(P<0.05),可能影响后期实验结果观察;(3)双侧精囊注射角叉菜胶较前列腺左右腹侧注射法效果更佳(P<0.05),但是增高了大鼠死亡率,并且出现了急性炎症反应;相比较前列腺左右腹侧注射法构建模型成功率高,局部多点注射法会弥补浸润范围偏小这一不足;(4)3%角叉菜胶生理盐水溶液较1%角叉菜胶生理盐水溶液建模效果更显著(P<0.05)。结论SD雄性大鼠比Wistar雄性大鼠对角叉菜胶更敏感;3%角叉菜胶生理盐水溶液在SD雄性大鼠前列腺左右腹侧叶适量多点局部注射,21 d后会成功构建稳定且理想的CNP模型。Objective To explore the influencing factors of carrageenan onchronic nonbacterial prostatitis(CNP)modelsin rats.Methods Adult male rats were injected with carrageenan to make models,and the effects of mouse species(SD rats and Wistar rats),injection sites(seminal vesicle andventral lobe of prostate)and injection concentration(1%and 3%)on the modeling were observed.The modeling effect was determined according to the results of HE stainingof prostate histopathological indicators.Results Compared with Wistar male rats,the results of HE staining in SD male prostate tissues were significantly higher(P<0.05).Spontaneous inflammation appeared in parts of normal Wistar male rats(P<0.05),which mightaffect the observation of later experimental results.The effect of bilateral seminal vesicle injection was better than that of left and right ventral prostatic injection(P<0.05),but it increased the mortalityof rats and acute inflammatory reaction occurred;compared with left and right ventral prostaticinjection,the successful rate of model construction was higher,and the local multi-point injection method could make up for the lackof small infiltration area.The modeling effect of 3%carrageenan saline solution was more significant than that of 1%carrageenan saline solution(P<0.05).Conclusions SD male rats are more sensitive to carrageenan than Wistar male rats.A stable and ideal CNP model will be successfully established after 21 days of local injection of 3%carrageenan saline solution into the left and right ventral lobe of the prostate of SD male rats.
