摘  要:目的应用网络药理学研究方法挖掘白花蛇舌草-半枝莲抗肿瘤的物质基础及其作用机制。方法通过中药系统药理学技术平台(TCMSP)数据库检索白花蛇舌草、半枝莲两味中药的主要活性成分,并进行靶点预测。通过TTD、Drug Bank和Gene Cards数据库对其作用靶点进行筛选得到与肿瘤相关的作用靶点。应用STRING平台构建蛋白-蛋白(PPI)互作网络模型,并使用Cytoscape软件对该网络进行拓扑学分析得到核心靶点。通过Metascape软件并对核心靶点进行GO-BP生物功能富集分析以及KEGG通路分析。结果经数据库分析白花蛇舌草中含有活性化合物7个、作用靶点282个;半枝莲中含有活性化合物29个、作用靶点428个,两味药中与肿瘤相关的作用靶点421个。GO-BP生物功能富集以及KEGG通路分析发现,两味中药主要通过激酶活性的调控、跨膜受体蛋白酪氨酸激酶信号通路、肽基丝氨酸磷酸化、细胞对氮化合物、有机环状化合物以及无机物的反应、阳离子稳态、磷脂酰肌醇介导的信号传导等生物学过程发挥其抗肿瘤作用,涉及到的肿瘤有前列腺癌、非小细胞肺癌、胰腺癌、膀胱癌、胶质瘤、慢性粒细胞白血病、小细胞肺癌、黑色素瘤、子宫内膜癌、急性粒细胞白血病、肾细胞癌、大肠癌和甲状腺癌等。结论该研究从多角度探讨了白花蛇舌草—半枝莲两味中药抗肿瘤的物质基础及其作用机制,为其临床和基础研究提供新的科学依据。Objective To explore the material basis and mechanism of anti-tumor effect of Hedyotis diffusa and Scutellaria barbata by network pharmacology.Methods The main active components of Herba Hedyotidis diffusae and Herba Scutellariae barbatae were retrieved through TCMSP database,and target prediction was carried out.According to TTD,Drug Bank and Gene Cards databases,the targets of action were screened to obtain tumor-related targets.The protein-protein(PPI)interaction network model was constructed by STRING platform,and the core target was obtained by topology analysis of the network using Cytoscape software.Through Metascape software,GO-BP biological function enrichment analysis and KEGG pathway analysis were carried out on the core targets.Results According to database analysis,Hedyotis diffusa contains 7 active compounds and 282 targets.Scutellaria barbata contains 29 active compounds,428 action targets and 421 tumor-related action targets in the two drugs.The GO-BP biological function enrichment and KEGG pathway analysis found that the two traditional Chinese medicines exert their anti-tumor effects mainly through biological processes such as kinase activity regulation,transmembrane receptor protein tyrosine kinase signal pathway,peptide serine phosphorylation,cell reactions to nitrogen compounds,organic cyclic compounds and inorganic substances,cation homeostasis,phosphatidylinositol mediated signal transduction,etc.The tumors involved include prostate cancer,non-small cell lung cancer,pancreatic cancer,bladder cancer,glioma,chronic myeloid leukemia,small cell lung cancer,melanoma,endometrial cancer,acute myeloid leukemia,renal cell cancer,colorectal cancer and thyroid cancer.Conclusion In this study,the material basis and mechanism of anti-tumor effect of Hedyotis diffusa and Scutellaria barbata were discussed from various angles,providing new scientific basis for its clinical and basic research.
