摘  要:目的研究橙皮苷对果糖-高脂饮食诱导的非酒精性脂肪性肝病(NAFLD)小鼠肝脏Toll样受体-4(TLR4)和Toll样受体-9(TLR9)表达的影响。方法60只昆明小鼠随机分为正常、模型、辛伐他汀、橙皮苷低、中和高剂量组,n=10,10%果糖水+高脂饲料诱导造模,后接受4周治疗,治疗结束后收集血清检测丙氨酸氨基转移酶(ALT)、天冬氨酸氨基转移酶(AST)、总胆固醇(TG)、游离脂肪酸(FFA)、白介素-1(IL-1)和肿瘤坏死因子(TNF-α)含量,收集肝脏苏木精伊红(HE)染色检测肝脏病理学,以及采用实时荧光定量PCR和Western Blotting检测肝脏组织Toll样受体-4(TLR-4)和TLR-9蛋白及m RNA表达。结果肝脏组织病理学可见正常组正常,而模型组则出现弥漫性脂肪颗粒浸润,与模型组相比,各治疗组均明显改善;与正常组相比,模型组AST、ALT、TG、FFA、IL-1和TNF-α均明显升高(P<0.05),与模型组相比,各治疗组上述指标均明显降低(P<0.05),此外橙皮苷高剂量组AST、ALT、TG和FFA与辛伐他汀组相比并没有差异,而IL-1和TNF-α则明显低于辛伐他汀组(P<0.05);与正常组相比,模型组TLR-4和TLR-9蛋白表达均明显升高(P<0.05),各治疗组则明显降低,此外橙皮苷高剂量TLR-4和TLR-9明显低于辛伐他汀组(P<0.05)。结论橙皮苷能够明显改善果糖和高脂饮食诱导的脂肪肝病肝功能和血脂,同时降低炎症刺激,其作用机制与其可下调肝脏中TLR-4和TLR-9表达有关。Objective To observe the affection of hesperidin on the non-alcoholic fatty liver disease(NAFLD)mice constructed by fructose+high-fat diet from Toll-like receptor-4(TLR4)and Toll-like receptor-9(TLR9)proteins expression.Methods 60 Kunming mice were divided into 6 groups via random number method,which were the normal,model,Simvastatin,and low-dose,medium-dose and high-dose of nobiletin group,n=10,all mice were received high-fat diet+10%fructose water for 8 weeks to build NAFLD model,after 6 weeks treatment,their blood were collected for test the aspartate aminotransferase(AST),alanine aminotransferase(ALT),total cholesterol(TG),free fatty acid(FFA),interleukin-1(IL-1)and tumor necrosis factor(TNF-α),then collected their liver to detect the liver pathology,meanwhile using real-time quantitative PCR and Western Blotting detect the expression of Toll-like receptor-4(TLR4)and Toll-like receptor-9(TLR9)m RNA and protein in liver tissue.Results The histopathology of the liver showed the slices was normal in the normal group,while in the model group showed diffuse fat granule infiltration,compared with the model group,each treatment group improved significantly.Compared with the normal group,the model group for AST,ALT,TG,FFA,IL-1 and TNF-αwere significantly elevated,compared with the model group,the above indicators were significantly lower in each treatment group,in addition,there was no difference between AST,ALT,TG and FFA in the high-dose hesperidin group compared with the simvastatin group.IL-1 and TNF-αwere significantly lower than the simvastatin group;Compared with the normal group,the expression of TLR-4 and TLR-9 protein and m RNA in the model group was significantly increased,and in the treatment group was significantly reduced,in addition in the high doses of nobiletin group,the TLR-4 and TLR-9 were significantly lower than the simvastatin group.Conclusion Hesperidin could significantly improve the liver function,blood lipids and reduce inflammation of NAFLD mice induced by fructose and high-fat diet,and
