摘 要:目的研究伏九贴膏的乙醇提取工艺。方法以延胡索乙素、麻黄碱、芥子碱含量及出膏率为综合评价指标,通过L9(34)正交试验考察乙醇浓度、乙醇用量、提取时间及提取次数对伏九贴膏醇提工艺的影响。采用HPLC测定 3 种指标成分的含量,延胡索乙素∶流动相甲醇-0.1%磷酸溶液(三乙胺调pH值至6.0)(50∶50),检测波长280 nm;麻黄碱∶流动相乙腈-1.5%十二烷基硫酸钠(53∶47),检测波长210 nm;芥子碱∶流动相乙腈-0.08mol/L磷酸二氢钾溶液(10∶90),检测波长326 nm。结果最佳提取工艺为60%乙醇,6倍量,回流提取2次,每次1 h。延胡索乙素、麻黄碱、芥子碱含量分别为0.296%,0.079%,1.97%,平均干膏率 16.23%。结论优选的醇提工艺稳定可行,适用于伏九贴膏的工业化生产。Objective:To optimize extraction technology of Fujiu transdermal patch.Method:With dry extract rate and contents of tetrahydropalmatine,ephedrine,sinapine as comprehensive index,orthogonal design was adopted to investigate effects of the amount and concentration of ethanol,extracting time and times on extraction technology.Contents of tetrahydropalmatine,ephedrine,sinapine were determined by HPLC,mobile phase of tetrahydropalmatine was methanol-0.1% phosphoric acid solution(adjust pH value to 6.0 by triethylamine)(50∶50),detection wavelength was 280 nm;mobile phase of ephedrine was acetonitrile-1.5% sodium lauryl sulfate solution(53∶47),detection wavelength was 210 nm;mobile phase of sinapine was acetonitrile-0.08 mol/L monopotassium phosphate solution(10∶90),detection wavelength was 326 nm.Result:Optimal extraction technology was extracted 2 times with 6 times,the amount of 60% ethanol for 1 h per time.Extraction amounts of tetrahydropalmatine,ephedrine,sinapine were 0.296%,0.079%,1.97%,dry extract rate was 16.23%.Conclusion:This optimized extraction technology is stable,feasible and suitable for industrial production of Fujiu transdermal patch.
