摘 要:目的:观察益气养阴法中药组方——抗纤合剂对博莱霉素诱导的肺纤维化大鼠肺组织中Smad3和Smad7蛋白质表达的影响,探讨抗纤合剂治疗肺纤维化的可能分子作用机制。方法:将健康雄性Wistar大鼠120只随机分为正常对照组、模型组、强的松组、抗纤合剂组。每组30只。除正常对照组,其他3组采用Szapiel SV经典博莱霉素致肺纤维化模型。模型建成后,强的松组以6.25 mg·kg-1的强的松溶液灌胃,抗纤合剂组以5 g·kg-1灌胃。正常对照组与模型组以等溶生理盐水灌胃,每组每只大鼠每日灌胃给药2次,观察大鼠一般行为状态,分别于7、14、28 d 3个时间点处死大鼠,取肺组织固定。每个时间点每组大鼠10只。分别观察3个时间点Smad3 mRNA、Smad7mRNA的蛋白表达的变化。结果:模型组在14 d时,Smad3的mRNA表达水平达到峰值,Smad7的mRNA表达水平达到低点。到28 d时,Smad3的mRNA表达水平有下降,Smad7的mRNA表达水平有上升。强的松组和抗纤合剂组大鼠肺组织中Smad3 mRNA表达水平随着时间的推移,逐渐下降,Smad7 mRNA表达水平逐渐升高。且从总体上与模型组相比,Smad3 mRNA表达水平显著降低,Smad7 mRNA表达水平显著升高,差异明显(P<0.05)。结论:抗纤合剂可以对TGF-β/Smad信号通路中Smad3、Smad7 mRNA的表达进行有效调控,通过下调BLM致肺纤维化大鼠Smad3的表达,促进Smad7表达的升高,抑制大鼠肺纤维化的形成,对肺纤维化起到一定的治疗作用。Objective:The purpose of the research is to investigate the effect of the traditional Chinese medicine prescription of the benefiting qi and nourishing yin method——the antifibrous mixture on the protein expression of Smad3 and Smad7 in bleomycin-induced pulmonary fibrosis in rats,and to discuss the possible molecular mechanism of antifibrous mixture in this process.Method:120 healthy Wistar male rats were randomly divided into control group,model group,prednisone group and antifibrous mixture group.Each group has 30 rats.Except the control group,Szapiel SV Classical Bleomycin was used to establish the pulmonary fibrosis model in the other 3 groups.When the model was established,the prednisone group was given a prednisone solution of 6.25 mg·kg-1 by gavage,and the antifibrous mixture group was given a antifibrous mixture solution of 5 g·kg-1 by gavage.At the same time,the control group and the model group were using the normal saline for gavage.All rats in different groups were intragastric administrated with drugs twice a day.We observed the general state of rats,and killed and sampled the rats at 7 days,14 days and 28 days respectively.The lung tissue was fixed.The protein expressions of Smad3 mRNA and Smad7 mRNA were observed respectively on day 7,14 and 28.Ten rats were taken from each group at each selected date to observe their protein expression changes of Smad3 mRNA,Smad7 and mRNA.Results:The expression of Smad3 mRNA in the model group reached the peak on the 14 th day while that of Smad7 hit the lowest point on the same day.By 28 days,the expression level of Smad3 decreased,while the expression level of Smad7 increased.The protein expression level of Smad3 in rats treated by gavage,and the antifibrous mixture gradually decreased with time while that of Smad7 mRNA had a completely inverse trend(P<0.05).Comparing to that in the model group,the expression of Smad3 in the lung tissue of the antifibrous group was significantly decreased and the expression of Smad7 was remarkably increased.Conclusion:The
