摘  要:目的通过超高效液相色谱-电化学法(UPLC-ECD)检测莲子中的去甲乌药碱。方法采用UPLC-ECD检测莲子中的去甲乌药碱,将77%的甲醇作为提取溶剂,以0.22μm的滤膜过滤提取液,行色谱分离,制备标准曲线,分析UPLC-ECD对莲子中(包括莲子心和去心莲子)的去甲乌药碱的测定情况。结果UPLC-ECD检测去甲乌药碱的标准曲线在实验浓度0.05~10 mg/L内表现出良好线性关系(R^(2)=0.9987),去甲乌药碱的检测限(LOD)为2.08μg/L,样品平均回收率、相对标准偏差(RSD)分别在94.6%~97.4%、2.9%~3.6%。与去心莲子相比,莲子心中去甲乌药碱含量明显较高。结论UPLC-ECD可检测莲子不同部位中的去甲乌药碱。Objective To detect higenamine in Lianzi(Nelumbinis Semen)by ultra performance liquid chromatography-electrochemical method(UPLC-ECD).Methods The higenamine in lotus seeds was detected by UPLC-ECD.77%methanol was applied as the extraction solvent,and the extracting solution was filtered with the 0.22μm filter membrane.The chromatographic separation was conducted to prepare standard curves.The determination situations of higenamine in Lianzi(Nelumbinis Semen)[Lianzixin(Nelumbinis Plumula),heart-loss lotus seeds]by UPLC-ECD were analyzed.Results The standard curves which described that UPLC-ECD detected higenamine showed good linear relationship(R^(2)=0.9987)within the experimental concentration 0.05-10 mg/L.The limit of detection(LOD)of higenamine was 2.08μg/L.The average recycle rate and relative standard deviation(RSD)of samples were 94.6%-97.4%and 2.9%-3.6%,respectively.Compared with heart-loss lotus seeds,the content of higenamine in lotus plumule was significantly higher.Conclusion UPLC-ECD can detect higenamine at different sites of lotus seeds.
