摘  要:目的探讨肺康益气颗粒对脂多糖(LPS)诱导的大鼠急性肺损伤(ALI)的保护作用及免疫调节作用,为肺炎恢复期的临床治疗提供理论依据。方法采用气管内滴注LPS(5 mg/kg)构建ALI模型,将大鼠随机分为正常组、假手术组、模型组、肺康益气颗粒低(2.7 g/kg)、中(5.4 g/kg)、高(10.8 g/kg)剂量组、六君子丸(1.62 g/kg)组和地塞米松片(1.08 mg/kg)组。给药7 d后处死动物,腹主动脉采血,EDTA抗凝管采血2 mL,测定血清中单核细胞和嗜中性粒细胞;大鼠肺组织匀浆液ELISA法测定肺组织中TNF-α、IL-6含量。采用腹腔注射环磷酰胺(50 mg/kg)构建免疫功能低下模型,观察小鼠溶血素生成和迟发超敏反应。结果与模型组比较,高剂量组单核细胞计数显著降低(P<0.05)。低、中、高剂量组TNF-α显著降低(P<0.01),高剂量组IL-6显著降低(P<0.01)。与模型组比较,高剂量组对环磷酰胺所致免疫功能低下小鼠血清溶血素的生成有明显增加的作用(P<0.05);中、高剂量组均有明显增强环磷酰胺所致免疫功能低下小鼠迟发型超敏反应的作用(P<0.05)。结论肺康益气颗粒能明显减轻LPS诱导的ALI大鼠肺水肿,缓解肺组织损伤及炎症反应,对急性肺损伤有一定的保护作用,且能提高机体的免疫功能,适用于肺炎恢复期肺脾气虚证的治疗。Objective To investigate the protective effect and immunomodulatory effect of Feikang Yiqi Granule(肺康益气颗粒)on lipopolysaccharide(LPS)-induced acute lung injury(ALI)in rats and provide theoretical basis for clinical treatment of pneumonia in the concomitant period.Methods ALI model was established by intratracheal infusion of LPS(5 mg/kg),and the rats were randomly divided into normal group,sham operation group,model group,Feikang Yiqi Granule low-dose(2.7 g/kg),medium-dose(5.4 g/kg)and high-dose(10.8 g/kg)groups,Liujunzi Pill(六君子丸)(1.62 g/kg)group and dexamethasone tablet(1.08 mg/kg)group.Blood samples were collected from abdominal aorta and 2 mL blood samples were collected by EDTA anticoagulant tube.Monocytes and neutrophils in serum were determined.The contents of TNF-αand IL-6 in lung tissue of rats were determined by homogenate ELISA.The immunodeficiency model was established by intraperitoneal injection of cyclophosphamide(50 mg/kg)to observe hemolysin production and the delayed hypersensitivity in mice.Results Compared with that of the model group,the monocyte count in the Feikang Yiqi Granule high-dose group was significantly decreased(P<0.05).TNF-αwas significantly decreased in Feikang Yiqi Granule low-dose,medium-dose and high-dose groups(P<0.01)and IL-6 was significantly decreased in Feikang Yiqi Granule high-dose group(P<0.01).Compared with that of the model group,the production of serum hemolysin in mice with immunodeficiency induced by cyclophosphamide in the Feikang Yiqi Granule high-dose group was significantly increased(P<0.05).Feikang Yiqi Granule medium-dose and high-dose groups significantly enhanced the delayed type hypersensitivity in mice with immunodeficiency induced by cyclophosphamide(P<0.05).Conclusion Feikang Yiqi Granule can significantly reduce LPS-induced pulmonary edema in ALI rats,relieve lung tissue damage and inflammatory response,and has a certain protective effect on acute lung injury.It can improve the immune function of the body,and is suitable for the t
