摘  要:目的探讨消癌解毒方及醇提物对肝癌SMMC-7721细胞增殖及P53、P21基因表达的影响。方法体外培养肝癌细胞,分为肝癌对照组、消癌解毒方组和醇提物组,不同剂量药物分别作用于肝癌细胞24 h、48 h和72 h后,CCK-8法检测各组细胞增殖活性,数码倒置显微镜观察细胞形态,实时定量PCR检测P53和P21基因表达情况。结果作用剂量上,22μg/mL消癌解毒方组、100μg/mL和150μg/mL醇提物组对细胞增殖的抑制作用优于其它剂量组;作用时间上,72 h抑制细胞增殖作用优于48 h和24 h;22μg/mL消癌解毒方组和100μg/mL醇提取物组对细胞形态影响显著;且明显提高P53和P21基因的表达。结论消癌解毒方及醇提物通过上调肝癌SMMC-7721细胞P53基因的表达,并激活其下游P21基因的表达,从而抑制肝癌细胞增殖可能是其抗肿瘤作用机理;而醇提物可能是该方发挥药效的有效作用部位。Objective To study the Influence of Xiaoai Jiedu Prescription and ethanol extracts on proliferation and expressions of P53 and P21 genes in hepatocarcinoma SMMC-7721 cell.Methods Hepatocarcinoma cells were cultured in vitro and were divided into liver cancer control group,Xiaoai Jiedu Prescription group and ethanol extracts group.Different doses of drugs were applied to hepatocarcinoma cells 24 hours,48 hours and 72 hours later,respectively.CCK-8 was used to detect cell proliferation activity.Inverted microscope was used to observe the cell morphology and real-time Q-PCR was used to detect the expressions of P53 and P21 genes.Results The inhibition effect of 22μg/mL Xiaoai Jiedu Prescription,100μg/mL and 150μg/mL ethanol extracts groups on cell proliferation was superior to that of the other groups.The inhibition effect of 72 hours on cell proliferation was better than that of 48 and 24 hours.Cell morphology was significantly affected and the expressions of P53 and P21 genes were significantly improved by 22μg/mL Xiaoai Jiedu Prescription and 100μg/mL ethanol extracts groups.Conclusion Inhibiting cell proliferation and increasing the expressions of P53 and P21 genes may be the anti-tumor mechanism of Xiaoai Jiedu Prescription and ethanol extracts.The ethanol extracts may be an effective part of this prescription.
