摘  要:目的探讨血小板凝胶(PG)治疗海水浸泡伤的效果。方法连续分3批次采用健康BALB/c小鼠心脏穿刺取血,提取富血小板血浆,通过血细胞分析仪评价富血小板血浆提取前后血细胞成分的变化;随后,以牛凝血酶和钙离子激活剂引发凝血连锁反应,制作血小板凝胶,并对其生长因子含量进行测定。选取健康雄性BALB/c小鼠15只,建立小鼠腹部开放伤创伤动物模型,随机分为3组:单纯创伤(腹部开放伤预处理)对照组(normal control treatment,NCT)、创伤合并海水浸泡(经海水浸泡60 min二次打击)对照组(seawater control treatment,SCT)及血小板凝胶治疗组(seawater platelet gel treatment,SPT),观察PG对小鼠创面愈合率及愈合时间的影响。结果3个批次所提取的富血小板血浆中血小板(platelet,PLT)提取率为(82.04±0.02)%,计数值为(464.67±31.18)×109/L,白细胞(white blood cell,WBC)、红细胞(red blood cell,RBC)总体残余率均<1%。所形成的PG提取液中血管内皮生长因子(vascular endothelial growth factor,VEGF)、表皮生长因子(epidermal growth factor,EGF)、血小板源性生长因子(platelet-derived growth factor,PDGF)的含量测定结果分别为(472.33±36.77)、(279.67±23.03)、(464.00±64.09)pg/ml。海水浸泡伤后,NCT组创伤愈合时间为(11.20±0.27)d、SCT组为(15.10±0.41)d;予以血小板凝胶治疗后,SPT组愈合时间为(12.30±0.44)d,3组的愈合时间差异有统计学意义(F=134.778,P<0.01);创面愈合率第5、7、10、12天,SPT组[分别为(55.02±2.76)%、(70.54±4.68)%、(92.76±1.10)%、100%]显著高于SCT组[分别为(49.91±1.23)%、(58.02±2.77)%、(69.89±5.74)%、(81.70±5.43)%],NCT组[分别为(55.19±1.34)%、(79.24±1.06)%、(95.18±1.02)%、100%]显著高于SCT组,3组的创面愈合率差异有统计学意义(F=76.742,P<0.01)。结论富PG治疗可缩短创面愈合时间,提高愈合率,促进小鼠海水浸泡伤后创面的修复。Objective To investigate the effect of platelet gel(PG)on seawater immersion wound.Methods The plate⁃let⁃rich plasma(PRP)was extracted by heart puncture of healthy mice continuously for three batches,and blood cell com⁃ponents were evaluated with a blood cell counter.Then,blood clotting linkage reaction was initiated by the bovine throm⁃bin and calcium ion activator to obtain PG.Levels of growth factors were determined by ELISA.Fifteen healthy female mice were selected and randomly divided into three groups to establish animal models for trauma of open abdominal injury;the simple wound control group(mice whose open wounds of the abdomen were treated,normal control treatment,NCT),trau⁃ma and seawater immersion group(mice whose open wounds of the abdomen were treated in combination with seawater im⁃mersion without PG treatment,SCT),and PG treatment group(mice whose open wounds of the abdomen were treated in combination with seawater immersion with PG treatment,SPT).The effect of PG on the wound healing rate of mice was monitored.Results The extraction rate of PLT in the extracted PRP was(82.04±0.02)%,the count value was(464.67±31.18)×109/L,and the overall residual rates of WBC and RBC were less than 1%.The vascular endothelial growth factor(VEGF),epidermal growth factor(EGF),and platelet⁃derived growth factor(PDGF)in the formed PG extract were deter⁃mined as(472.33±36.77),(279.67±23.03),and(464.00±64.09)pg/ml,respectively.The time taken by wound healing was(11.20±0.27)days and(15.10±0.41)days for NCT and SCT respectively.After seawater immersion injury,the time taken by wound healing in the SPT group was(12.30±0.44)days and the comparison between the three groups was P<0.01.The re⁃epithelialization rate of the SPT group was(55.02±2.76)%,(70.54±4.68)%,(92.76±1.10)%and 100%respective⁃ly on days 5,7,10 and 12,compared with(49.91±1.23)%,(58.02±2.77)%,(69.89±5.74)%,and(81.70±5.43)%in the SCT group,and(55.19±1.34)%,(79.24±1.06)%,(95.18±1.02)%,and 100%in the NCT group,respectively.Th
