摘 要:目的:比较RNA恒温扩增-金探针层析法和荧光PCR法检测肺炎支原体的结果,探讨前者在肺炎支原体检测中的临床应用价值。方法:采用整体抽样的方法收集344例临床咽拭子标本,分别用RNA恒温扩增-金探针层析法和荧光PCR法进行检测,以荧光PCR法所得结果为参比,对检测结果进行统计分析。结果:RNA恒温扩增-金探针层析法阳性检出率13.08%,荧光PCR法阳性检出率12.79%,差异无统计学意义(χ2=0.0129,P>0.05)。RNA恒温扩增-金探针层析法与荧光PCR法相比阳性符合率为97.73%,阴性符合率为99.33%,总符合率为99.13%,一致性系数(Kappa值)为0.9613。结论:二者总符合率高,有相同的检出能力。在此基础上,RNA恒温扩增-金探针层析法作为一种创新方法,还具有操作简单、设备要求不高、结果判读明了等优点,因此该方法在肺炎支原体检测中有很强的临床应用价值。Objective:To compare the results of mycoplasma pneumoniae detection between RNA thermo-static amplification-colloidal gold method and fluorescence PCR,and to explore the clinical application of the former in the detection of mycoplasma pneumoniae.Methods:A total of 344 clinical pharyngeal swabs were collected by the whole sampling method.The samples were detected by RNA thermostat-ic amplification-colloidal gold method and fluorescence PCR method respectively.The results ob-tained by fluorescence PCR method were used as the reference for statistical analysis of the test re-sults.Results:The positive rate of RNA thermostatic amplification-colloidal gold method was13.08%,and that of fluorescence PCR was 12.79%,the difference was not statistically significant(χ2=0.0129,P>0.05).According to the results of comparing with fluorescence PCR,the positive co-incidence rate of RNA thermostatic amplification-colloidal gold method was 97.73%,the negative co-incidence rate was 99.33%,the total coincidence rate was 99.13%,and the Kappa value was0.9613.Conclusion:The two methods have high coincidence rate and the same detection ability.On this basis,the RNA thermostatic amplification-colloidal gold method,as an innovative method,has the advantages of simple operation,low equipment requirements,and clear interpretation of results.Therefore,this method has a strong clinical application value in the detection of mycoplasma pneumoniae.
