摘 要:百合中富含多糖类、生物碱类、甾体皂苷类、黄酮类以及酚类化合物等多种化学成分,具有止咳平喘、抗氧化、抗抑郁、抗肿瘤、抗真菌、降血糖和免疫调节等多种药理作用,被广泛用于临床中药配方和中成药。道地百合具有明显的地域性,不良商家常以其它产区的百合冒充道地百合牟利,因此需发展操作简单且能快速对百合进行品质鉴定和产地溯源的技术。本研究采取湖南平江、株洲和龙山以及江西万载4个产地的百合鳞茎样本共60份,经过烫片、热风烘干、粉碎和过筛等前处理,然后扫描其固体粉末荧光光谱,最后利用主成分分析判别方法(Principal component analysis-linear discriminant analysis,PCA-LDA)和偏最小二乘判别分析方法(Partial least squares discriminant analysis,PLS-DA)构建百合产地判别模型。结果表明,基于百合粉末荧光光谱的判别模型能够对不同产地的百合样本进行准确的溯源分析。PCA-LDA对训练集和测试集的分类准确率(Correct classification rate,CCR)分别为93.6%和95.6%,PLS-DA对训练集和测试集的CCR分别为98.3%和93.3%。基于百合粉末荧光光谱的分类结果与基于百合提取液的三维荧光光谱的分类结果无显著性差异。本方法操作简单、检测速度快,并且不损耗样本,不需要使用有机溶剂,是一种绿色环保的检测方法,适合于大批量百合样品的质控检测。Lily has certain functions of relieving cough and asthma,antioxidation,anti-depression,anti-tumor,anti-fungi,hypoglycemic effect,and immunoregulation as well.Due to the diversity of the geographical origin of lily and the resulting apparent differences in their quality,lily products in different levels of quality are intermingled in the market.Some sellers even sell shoddy lily at high price.Therefore,methods capable of rapid evaluating quality of lilies and tracing their geographical origins are highly desired.In this work,a total of 60 samples of lily bulbs harvested from four different regions in China(i.e.,Pingjiang,Zhuzhou,Longshan and Wanzai)were first undergone treatments of scalding,hot-air drying,crushing and sieving.The fluorescence spectra of the obtained powder samples were then measured and used to build discriminant models for determining geographical origins of lily by principal component analysis-linear discriminant analysis(PPCA-LDA)and partial least squares discriminant analysis(PLS-DA).The results demonstrated that the discriminant models based on the fluorescence spectra of lily powder samples could accurately identify the geographical origins of lily samples.The correct classification rate(CCR)obtained by PCA-LDA for training set and test set were 93.6%and 95.6%,respectively.The corresponding values obtained by PLS-DA were 98.3%and 93.3%,respectively.There was no significant difference between the classification results based on the fluorescence spectra of lily powder samples and those based on the three-dimensional fluorescence spectra of lily extracts.The proposed method showed the merits of simple implementation,rapid detection,non-destructiveness,solvent-free,and environmentally-friendly,and therefore suitable for quality control of a large number of lily products.
