摘  要:乙烯受体基因ETR1是乙烯信号通路上的关键基因,在调控植物的生长发育与抵抗生物胁迫方面扮演着十分重要的角色,据此推断丹参(Salvia miltiorrhiza)ETR1是调控其生长发育的关键因子,并能直接影响其品质,但目前有关丹参乙烯受体基因的相关研究仍未见报道。本研究利用芝麻的ETR1基因序列在丹参转录组数据库里比对获得了一条高度同源的序列,通过设计特异性引物克隆获得长为2223 bp的丹参ETR1序列(命名为SmETR1),生物信息学分析显示SmETR1是含跨膜结构的非分泌型疏水蛋白,定位于内质网,且含有GAF、HisKA、HATPase_c、Response_reg等乙烯受体ETR1的典型结构域,具有丝氨酸为主、苏氨酸为辅对乙烯信号转导进行磷酸化的修饰与调控位点,SmETR1启动子分析显示其含有与分生组织表达、响应茉莉酸甲酯信号、响应脱落酸信号及厌氧诱导等作用元件,这些元件可能也参与了由乙烯介导的信号通路。SmETR1在丹参叶内具有较高的表达水平。本研究为丹参乙烯受体基因功能研究奠定基础,同时可为利用基因策略进行丹参分子育种研究提供可参考的理论依据。Ethylene receptor gene(ETR1),which is a key gene of ethylene signaling pathway,plays a very important role in plant growth and biological stress.It is inferred that ETR1 is a key factor in regulating the growth and development of Salvia miltiorrhiza,and it can also directly affect its quality.However,the research on ethylene receptor gene in S.miltiorrhiza has not been reported.Here,a highly homologous sequence was obtained from S.miltiorrhiza transcriptome database by comparing the sequences of ETR1 gene in Sesamum indicum L..The 2223 bp sequence(named SmETR1)was cloned.The SmETR1 bioinformatics analysis showed that it was the non-secreted hydrophobic protein,which locates in endoplasmic reticulum.SmETR1 contains the typical domains of ethylene receptor ETR1,such as GAF domain,HisKA domain,HATpase_c domain and Response_reg domain.The SmETR1 contains Ser-based,Thr-assisted phosphorylation-modulatory which regulate ethylene signaling.The upstream promoter analysis showed it contains meristem expression,methyl jasmonate signal,abscisic acid signal and anaerobic induction elements.These elements may also participate in ethylene mediated signaling pathway.The expression level of SmETR1 was higher in leaves than other organs of S.Miltiorrhiza.These results put a foundation for the functional research of ethylene receptor gene in S.miltiorrhiza.These analyses also provide theoretical basis for molecular breeding of S.miltiorrhiza by gene strategy.
