摘  要:目的筛选在尖锐湿疣(condyloma acuminatum,CA)组织中差异表达的microRNA(miRNA),探究其在CA发病机制中的作用.方法采用RNAseq方法筛选在CA组织中差异表达的miRNA,运用生物信息学对差异表达的miRNA进行靶基因预测和功能分析.结果RNAseq方法筛选出CA组织中差异表达的miRNA 44个,其中31个miRNA上调,13个miRNA下调.对差异表达的miRNA进行靶基因预测,44个差异表达的miRNA的靶基因共有51928个.对这些靶基因进行GO和KEGG富集分析,结果显示其主要涉及丝裂原活化蛋白激酶(MAPK)信号转导通路、趋化因子信号通路、ErbB信号通路,内吞作用以及多种肿瘤的发生、发展,细胞黏附等生物学过程.结论CA组织中存在差异表达的miRNA,这些差异表达的miRNA调控的靶基因及其基因功能可能与CA的发病密切相关.Objective To explore the differential expression of microRNA(miRNA)profile in condyloma acuminatum(CA)tissue and to explore the role of miRNA in the pathogenesis of CA.Methods RNAseq method was used to detect the differential expression of miRNA in CA tissues,and bioinformatics was used to predict the target genes of differential expression of miRNA and analyze the functions.Results A total of 44 differential miRNAs were screened out by the RNAseq method,among which 31 miRNAs were up-regulated and 13 miRNAs were down-regulated.The target genes of the differential miRNAs were predicted,and there were 51928 target genes of 44 differential miRNAs.GO and KEGG enrichment analysis of these target genes showed that they were mainly involved in mitogen-activated protein kinase(MAPK)signal transduction pathway,chemokine signaling pathway,ErbB signaling pathway,endocytosis,the occurrence and development of various tumors,cell adhesion and other biological processes.Conclusion There are differential expressions of miRNAs in CA tissues,and the target genes regulated by these differentially expressed miRNAs and their gene functions may be clolyse related to the pathogenesis of CA.
