摘 要:目的探讨促卵泡刺激素(FSH)在玻璃化冻存全程保护性干预对小鼠卵巢组织形态结构及碱性成纤维细胞生长因子(bFGF)蛋白表达的影响。方法将18枚卵巢组织随机分成新鲜对照组(CG)、玻璃化冻存对照组(VCG)、0.3IU/mL FSH全程干预玻璃化冻存组(OG-FSH),每组6枚卵巢组织。观察玻璃化冻存后各组卵巢组织的形态结构改变,采用免疫组织化学法及western-blot技术观察并分析各组bFGF蛋白的表达情况。结果0.3IU/mL FSH全程干预玻璃化冻存组(OG-FSH)小鼠卵巢组织形态结构保持效果好,正常卵泡百分比显著高于玻璃化冻存对照组(VCG),P<0.05;0.3IU/mL FSH全程干预玻璃化冻存组(OG-FSH),bFGF蛋白表达量显著高于玻璃化冻存对照组(VCG),P<0.05。结论FSH在小鼠卵巢玻璃化冻存过程中发挥积极的卵巢形态结构保护效果,同时促进bFGF蛋白的表达,可减少冻存过程中冰晶形成、卵巢早衰等不利因素发生,提高冻存效率。Objective To investigate the effect of the expression ofbFGF after FSH interpose in the whole process of vitrification.Methods Eighteen ovarian tissues were randomly divided into three groups.The first group is fresh control group(CG),the second group is vitrified cryopreservation control group(VCG),and the last group isvitrification group intervention by 0.3IU/mL FSH(OG-FSH).Each group has 6 pieces of ovarian tissues.Then to observation the morphological and structural changes of ovarian tissues in each group after vitrification,and to observe and analyze the expression of bFGF protein in each groupby immunohistochemistry and western blot.Results The result from HE staining show that the morphological structure of mouse ovarian tissue keepsgetting better in OG-FSH group,the percentage of normal follicles in VCG group was significantly lower than CG group and VCG group(P<0.05);The expression of bFGF protein in OG-FSH group was significantly higher than VCG group(P<0.05).Conclusion FSH plays a positive role to protect the morphology and structure of mouse ovaries after vitrification,and FSH can promotes the expression of bFGF protein,to reduce the formation of ice crystals,for improve the vitrification efficiency.
