摘 要:甜菜是我国北方(包括西部)地区的重要糖料作物,当前我国甜菜种植面积大概有20万亩.连年的甜菜种植,出现的根腐病会直接导致甜菜含糖量降低,产量减少,严重时甚至绝产.生物防治甜菜根腐病作为环境友好型防治措施,正在被广泛关注.针对目前尚缺乏对甜菜根际土壤中细菌群落结构的研究,本研究采用稀释涂平板分离方法,将采自内蒙古乌兰察布市集宁区健康甜菜的根际土样中菌株进行分离;通过16S rRNA的PCR扩增测序,测出土壤中细菌;并利用进化树显示出其亲缘关系.共分离出30株细菌,链霉菌属15株,芽孢杆菌属9株,节杆菌属1株,叶杆菌属1株,绿脓杆菌属1株,冢村氏菌属1株,贪噬菌属1株,黄杆菌属1株.冢村氏菌属细菌菌落总数(CFU)最多;链霉菌属菌株数量最多.希望能为生物防治甜菜根腐病提供一定依据,从而达到甜菜增产、农民增收的目标.Sugar beet is an important sugar crop in northern China(including western China).At present,its planting area in China has reached about 200 thousand mu.The root rot disease,usually taking place after years’of its planting,will lead to the decrease of sugar content,the reduction of yield or even no yield at all.Biological control,as an environmental-friendly control,has drawn wide concern.For the lack of research on the structure of bacterial community in the rhizosphere of sugar beet till now,the article adopts the dilution plating procedure and 16S rRNA sequencing of amplicon sequencing,detects bacteria in the healthy sugar beet’s rhizosphere samples in Ji’ning area,Wulanchabu,Inner Mongolia.Then the bacteria are isolated effectively,strains are cultivated and at last phylogenetic tree is mapped to show their genetic relationship.In total,thirty strains are isolated.Among these thirty strains,fifteen strains belong to Streptomyces,nine strains belong to Bacillus,one strain belongs to Arthrobacter,another strain belongs to Phyllobacterium,one strain belongs to Pseudomonas,one strain belongs to Tsukamurella,one strain belongs to Variovorax,and one strain belongs to Microbacterium.The CFU of Tsukamurella is the most,and the number of the strains of Streptomyces is the most.It is hoped that this article can provide certain basis for the biological control of sugar beets’root rot disease,thus to achieve the goal of yield increase for sugar beet and income increase for farmers.
