‘开远’滇杨的多倍体诱导及鉴定

摘　　要：在组织培养条件下,以'开远'滇杨幼嫩茎段为实验材料,用秋水仙素作为诱导剂,对'开远'滇杨进行多倍体诱导,比较不同预培养时间、 不同处理浓度以及不同处理时间下秋水仙素的诱导效果.结果表明:'开远'滇杨幼嫩茎段在分化培养基上预培养3d后,再在添加有40mg/L秋水仙素的分化培养基中处理3d,其诱导效果最佳,变异率可达50.79%.'开远'滇杨多倍性材料'同质化'处理的最佳次数为6次;对所得的多倍体材料进行流式细胞术检测,其倍性为四倍.The colchicine was used to induce female diploid Populus yunnanensis‘Kaiyuan’based on the tissue culture,polyploid induction was carried out on the cultivar,and the induction effects of colchicine were compared under different pre-culture time,different treatment concentrations and different treatment time.The results showed that optimal induction effect would be obtained when the young stem of P.yunnanensis‘Kaiyuan’were pre-cultured on differentiation medium for 3 days,then treated with the differentiation medium supplemented with 40mg/L colchicine for 3 days.The variation rate would be 50.79%.The number of optimal homogenization of the polyploidy materials was six times.Based on the analysis of flow cytometry,the ploid of polyploidy materials was tetraploid.
