摘  要:为建立中山杉组培无菌体系,以枝条类型、0.2%HgCl2处理时间、80%多菌灵及PVP为试验因素,采用正交试验设计L9(34),试验数据采用多指标综合平衡法进行分析,结果表明:枝条类型为各试验指标的主要因素,且影响都极显著;80%多菌灵对各试验指标的影响小,为不重要因素予以剔除,得出采用半木质化枝条作为外植体,0.2%HgCl2处理10min,MS培养基中加入PVP 0.5g/L可获得较为理想的中山杉组培无菌体系.In order to establishment of sterile culture of Taxodium hybrid‘Zhongshansa’,the test was designed by orthogonal of L9(34),with branch type,0.2%HgCl2 treatment time,80%carbendazim and PVP as experimental factors.The test data are analyzed by multi-index comprehensive balance method.The results showed that the branch type was the main factor of each index,and the influence was very significant.The effect of 80%carbendazim on each test index was small,which was considered to be an unimportant factor and could be eliminated.It was concluded that the ideal aseptic system of Taxodium hybrid‘Zhongshansa’could be obtained by using semi-lignified branches as explants,treating with 0.2%HgCl2 10 minutes and adding PVP 0.5g/L into MS medium.
