摘 要:目的:研究颈椎病大鼠颈脊髓中Caspase-3的表达水平及其在神经细胞凋亡中的作用.方法:选取65只4个月龄SD大鼠,破坏颈椎后柱稳定性后分别饲养3个月、5个月,运动功能评估用斜板试验;HE染色在形态学上检测颈椎间盘组织的变化;Western Blot和免疫组织化学染色在蛋白水平检测颈脊髓中Caspase-3的表达水平;大鼠颈脊髓神经细胞的凋亡用TUNEL法特异性检测,上述检测结果组间比较,并进行统计学分析处理.结果:运动功能的评价应用斜板试验,3个月对照组与观察组斜板角度为69.8° ±4.6°及45.5° ±3.7°,5个月对照组与观察组斜板角度为70.2° ±6.3°及43.6° ±4.5°;颈椎间盘C4-5、C5-6 Miyan-moto分级评估退变程度,3个月对照组与观察组分别为1.28±0.36及2.75±0.63,5个月对照组与观察组分别为1.43±0.42及4.89±0.71.经斜板试验及组织学观察,对照组与观察组间均有统计学意义,P<0.01.颈椎病大鼠颈脊髓Caspase-3的阳性细胞数3个月对照组与观察组分别为3.21±1.02及15.25±3.48,5个月对照组与观察组分别为4.27±2.83及28.41±4.89;Western Blot检测Caspase-3在不同组间的表达水平,3个月、5个月观察组间有统计学意义,P<0.01.结论:通过破坏大鼠颈椎后柱稳定可成功建立颈椎病模型.Caspase-3作为细胞凋亡过程中的重要酶之一,能够影响慢性脊髓损伤后细胞的凋亡,并且Caspase-3随着颈椎退变程度的增强而上调.Objective:To study expression of Caspase-3 and the neuron apoptosis in cervical cord of rats with cervical spondylosis.Methods:Sixty-five four month year old SD rats were operated with destroying cervical posterior column,3 months and 5 months later,the rats were detected motor function by oblique board test,cervical disc morphological changes were observed by HE dyeing.Then Caspase-3 expressions were tested by immunohistochemistry and Western Blot methods,neuron apoptosis were detected by TUNEL method,compared with control groups,data were calculated by statistical methods.Results:The motor function by oblique board test of 3 and 5 months(Control groups VS model groups)were 69.8±4.6 VS 45.5±3.7 and 70.2±6.3 VS 43.6±4.5 respectively.The degenerated degree of cervical disc morphological changes were evaluated by Miyanmoto grade,the results of 3 months and 5 months(Control groups VS model groups)were respectively that:1.43±0.42 VS 2.25±0.48,and 1.38±0.57 VS 4.22±0.69.There were significant difference between control and model groups,P<0.01.The positive cells of Caspase-3 expressions in cervical cord were 3.21±1.02 VS 15.25±3.48 and 4.27±2.83 VS 28.41±4.89 in 3 and 5 months of control and model groups.Western Blot detected the expression of Caspase-3 in control and model groups,P<0.05.Conclusion:The successful cervical spondylosis models could be established by destroying cervical posterior column.Caspase-3 as one of important enzyme during apoptosis,mediate neuron apoptosis in chronic spinal cord injury,and its increased expression gradually while aggravated degree of degenerated cervical disc.
