摘　　要：Objective: to observe the application effect of smear staining method in microbiological examination. Methods: 120 samples from our laboratory were selected as the research object for bacterial culture, and smear examination was performed before and after the operation. After the corresponding operation of bacterial culture was completed, the specific conditions of microorganism in smear examination were observed, and the actual conditions that the original samples were consistent with the smear examination after bacterial culture were counted, and the specific application effect of smear examination was analyzed. Results: through experimental observation, 106 samples of all original samples passed the test for the existence of microorganisms, mainly including fungi, gram-positive bacteria and gram-negative bacteria, among which gram-negative bacteria accounted for the highest proportion;Comparing the detection of microorganisms in the original specimen with the smear examination after the bacterial culture treatment, the urine, ascites, sputum, pus and feces were 80.00%, 75.00%, 80.00%, 58.33% and 40.00%, respectively. Conclusion: the direct smear examination of the original specimen and the smear examination after bacterial culture have a relatively high coincidence rate, which can make the type of original bacterial infection clearer and has certain practical application value.
