摘  要:目的观察血管紧张素(1-7)[Ang(1-7)]对血管紧张素Ⅱ(AngⅡ)诱导血管内皮细胞表达组织因子(TF)的影响。方法细胞培养采用RPMI-1640完全培养基:一期凝固法测总的促凝活性(PCA);RT-PCR方法检测TF mRNA。结果(1)不同浓度的AngⅡ(10-10~10-6mol.L-1)促进细胞TF活性和TF mRNA的表达,并具有明显的量效关系(r=0.9631,P<0.05);(2)单用Ang(1-7)(10-10~10-7mol.L-1)不能抑制细胞的TF活性(P>0.05);(3)在给予AngⅡ之前30 min用Ang(1-7)(10-10~10-7mol.L-1)预处理细胞,Ang(1-7)可呈剂量依赖式地抑制AngⅡ(10-7mol.L-1)对TF表达的刺激作用(r=0.9860,P<0.05),其在10-7mol.L-1时抑制作用达到最强。结论Ang(1-7)可抑制AngⅡ诱导的ECV304细胞表达TF,这一作用是在mRNA水平上实现的。Objective The purpose of this study is to observe the effects of angiotensin(I-7) [Ang (1-7)] on the expression of tissue factor(TF) induced by angiotensin Ⅱ(Ang Ⅱ) in human umbilical vein endoth elium derived cell. Methods TF activity was determined with one-stage clotting assay measuring total cellular procoagulant activity(PCA).TF mRNA was examined by semi-quantitative reverse transcription polymerase chain reaction(RT-PCR). Results (I)A gradual increase in PCA(r=0.9631,P<0.05) was observed in cell stimulatived with increasing concentration orang 11(10-10-10-6 mol/L).(2) Ang(1-7)(1010-10-7 mol/L) alone could not inhibit thc expression of TF.(P>0.05).(3) When pretreated with Ang(1-7)(10-1010-7 mol/L),Ang(1-7) could inhibit the expression of TF activity and mRNA induced by Ang Ⅱ(r=0.9860,P<0.05),and 10-7mol/L was the strongest concentration. Conclusion The present data suggests that Ang Ⅱ can induce the expression of TF in vascular endothelial cells and Ang(1-7) inhibited this effect at mRNA level.
