兔颈总动脉损伤后普罗布考对血管再狭窄的抑制作用

摘　　要：目的研究普罗布考减轻动脉成形术后再狭窄的作用。方法将30只新西兰大白兔随机分为普罗布考治疗组和对照组各15只。以球囊损伤颈总动脉,用高胆固醇食物饲养8周后切取颈总动脉,HE染色观察血管管腔面积和内膜面积的变化。采用明胶酶谱法和酶法测定动物血清中基质金属蛋白酶2的酶原(pro-MMP-2)、基质金属蛋白酶2(MMP-2)及血清胆固醇(TC)、低密度脂蛋白胆固醇(LDL-C)、甘油三酯(TG)、高密度脂蛋白胆固醇(HDL-C)的水平。结果普罗布考组TC、LDL-C、TG和HDL-C水平较对照组低,但差异无显著性(P>0.05);普罗布考组pro-MP-2、MMP-2水平较对照组低,两组比较均具有显著性差异(P<0.05);普罗布考组颈总动脉新生内膜面积、最大内膜厚度明显低于对照组,两者比较差异具有显著性(P<0.05);而普罗布考组颈总动脉管腔面积明显高于对照组后,两者比较差异具有显著性(P<0.05)。结论普罗布考可通过抑制MMPs的途径减轻兔颈总动脉成形术后再狭窄的形成。Objective To investigate the effect of Probucol on restenosis after carotid artery balloon injury.Methods 30 New Zealand White rabbits were randomly assigned to Probucol group(n=15)and control(n=15).Carotid artery tissues were harvested at weeks 8 after injury by balloon.During 8 weeks the rabbits were fed with hypercholesterolaemic diet.The areas of lumen of blood vessel and intima were observed by HE staining.The levels of serum pro-MMP-2 and MMP-2 were detected by zymograph and total cholesterol(TC),triglyceride(TG),high density lipoprotein cholesterol(HDL-C),low density lipoprotein cholesterol(LDL-C) were measured by enzyme method. Results Serum levels of TC,TG,HDL-C,LDL-C,pro-MMP-2 and MMP-2 in probucol group were lower than those in control,respectively;the expression levels of TC,TG,HDL-C and LDL-C in serum had no significant difference between two groups(P>0.05);there were obvious differences of pro-MMP-2,MMP-2 between two groups(P<0.05);there was a significant decrease in intimal area,intimal thickness and a significant increase in luminal area compared control with Probucol group(P<0.05).Conclusion Probucol can prevent restenosis after rabbits carotid artery balloon injury by inhibiting MMPs.
